Effect of ultraviolet light, methyl methanesulfonate and ionizing radiation on the genotoxic response and apoptosis of mouse fibroblasts lacking c-Fo…
2001
c-Fos and p53 are DNA damage-inducible proteins that are involved in gene regulation, cell cycle checkpoint control and cell proliferation following exposure to genotoxic agents. To investigate comparatively the role of c-Fos and p53 in the maintenance of genomic stability and the induction of apoptosis, we generated mouse fibroblast cell lines from knockout mice deficient for either c-fos (fos -/-) or p53 (p53-/-) or for both gene products (fosp53-/-). The sensitivity of these established cell lines was compared with the corresponding wild-type cells as to the cytotoxic, clastogenic and apoptosis-inducing effects of ultraviolet (UV-C) light and methyl methanesulfonate (MMS). Additionally, …
On Cancer Cell Cycle and Universal Apoptosis Parameters Signaling Unravelled In Silico
2010
Here, cell cycle in higher eukaryotes and their molecular networks signals both in G1/S and G2/M transitions are in silico replicated. Systems control theory is employed to design multi-nestled digital layers to simulate protein-to- protein activation and inhibition in the cancer cell cycle dynamics in presence of damaged genome. Sequencing and controlling the digital process of four micro-scale species networks (p53/Mdm2/DNA damage; p21mRNA/cyclin-CDK complex; CDK/CDC25/wee1/SKP2/APC/CKI and apoptosis target genes system) paved the way for unravelling the participants and their by-products having the task to execute (or not) cell death. The results of the proposed cell digital multi-layers…
Silymarin and Cancer: A Dual Strategy in Both in Chemoprevention and Chemosensitivity
2020
Silymarin extracted from milk thistle consisting of flavonolignan silybin has shown chemopreventive and chemosensitizing activity against various cancers. The present review summarizes the current knowledge on the potential targets of silymarin against various cancers. Silymarin may play on the system of xenobiotics, metabolizing enzymes (phase I and phase II) to protect normal cells against various toxic molecules or to protect against deleterious effects of chemotherapeutic agents on normal cells. Furthermore, silymarin and its main bioactive compounds inhibit organic anion transporters (OAT) and ATP-binding cassettes (ABC) transporters, thus contributing to counteracting potential chemor…
Bioinformatic and experimental fishing for artemisinin-interacting proteins from human nasopharyngeal cancer cells.
2012
Determining interacting cellular partners of drugs by chemical proteomic techniques is complex and tedious. Most approaches rely on activity-based probe profiling and compound-centric chemical proteomics. The anti-malarial artemisinin also exerts profound anti-cancer activity, but the mechanisms of action are incompletely understood. In the present investigation, we present a novel approach to identify artemisinin-interacting target proteins. Our approach overcomes usual problems in traditional fishing procedures, because the drug was attached to a surface without further chemical modification. The proteins identified effect among others, cell cycle arrest, apoptosis, inhibition of angiogen…
Piclamilast inhibits the pro-apoptotic and anti-proliferative responses of A549 cells exposed to H(2)O(2) via mechanisms involving AP-1 activation.
2012
Reactive oxygen species (ROS) are involved in the pathogenesis of many inflammatory diseases such as chronic obstructive pulmonary disease (COPD). They can alter the expression of genes involved in cellular damage by activating transcription factors, including the NF-κB and the activator protein 1 (AP-1). Phosphodiesterase type 4 (PDE4) inhibitors have anti-inflammatory and antioxidant effects, as described in in vivo and in vitro COPD models. This study analysed the effects of piclamilast, a selective PDE4 inhibitor, on modulating the global gene expression profile in A549 cells exposed to H(2)O(2).Changes in gene expression were analysed using high-density Affymetrix microarrays and valid…
Psychometric Properties of Two Brief Versions of the Hopkins Symptom Checklist: HSCL-5 and HSCL-10
2019
The Hopkins Symptom Checklist–25 (HSCL-25) is a widely applied measure of depression and anxiety. The present study examines two of its short forms—the HSCL-5 and HSCL-10, which have been proposed by previous research—in a representative sample of the German general population. To this end, we conducted exploratory and confirmatory analysis on two subsamples ( n = 1,246 and n = 1,216). Our results suggest that, compared with the HSCL-25, both short forms represent economical ways of assessing depression and anxiety. Model fit was good and correlations with established measures demonstrate convergent validity. Both HSCL short forms are strongly invariant across sex, and we found evidence fo…
Systematic review and critical appraisal of Childhood Trauma Questionnaire - Short Form (CTQ-SF).
2021
Abstract Background Child maltreatment is a complex and multidimensional construct that encompasses a great number of risk factors. The Childhood Trauma Questionnaire — Short Form, one of the most widely used and validated instruments to assess childhood maltreatment in the past ten years, is a retrospective instrument that assesses several types of childhood abuse and maltreatment which is divided into five dimensions. Objective The objectives of this systematic review are to critically appraise, compare, and summarize the methodological quality and psychometric properties of published research articles validating the Childhood Trauma Questionnaire — Short Form utilizing the COSMIN checkli…
Types of Offense among the Cleckley Psychopaths
1994
The Cleckley psychopath is superficially charming and convincing in his behavior but also unreliable and callous. These personality characteristics imply that he/she may be prone to deceive and manipulate others. In the area of criminality, this may mean apropensity to fraud-like offenses. The present paper tested the argument by comparing types of offense committed by psychopaths and nonpsychopaths. The Psychopathy Checklist was used for psychopathy assessments. In a sample of 92 male offenders, the percentage of the Cleckley psychopaths convicted for fraud-like offenses was higher than among the other offenders. The results were discussed with respect to the problem of circular conclusio…
Subjective symptomatology of asthma: validation of the asthma symptom checklist in an outpatient Spanish population.
1997
The objective of this study was to validate the Asthma Symptom Checklist (ASC) so that it could be reliably used to assess the subjective symptomatology of asthma attacks in our context. Subjective symptomatology of asthma was examined in a group of 100 adult Spanish outpatients (57 women, 43 men; 17-69 years of age) with asthma. All of them completed the modified version of the ASC as well as questionnaires of depression, anxiety, and self-management of asthma (self-efficacy expectancies and health care utilization). Data about duration and severity of asthma, as well as dyspnea and %FEV1, were also recorded. The highest reliability Cronbach alpha indexes were for the panic-fear and fatigu…
