Patterning of Suitable Structures for the Investigation of the Josephson Effect in YBa2Cu3O7/PrBa2Cu3O7 Superlattices
1996
The aim of our work is to investigate the Josephson effect in YBa 2 Cu 3 O 7-δ /PrBa 2 Cu 3 O 7-δ superlattices. In the case of current flow along c axis direction the superlattice forms an array of artificial Josephson junctions where the YBa 2 Cu 3 O 7-δ layers are the superconducting electrodes which are separated but Josephson coupled by the PrBa 2 Cu 3 O 7-δ sheets. In this paper we report on the preparation, characterisation and patterning of the superlattices into suitable structures via standard photolithograpy and lift-off technique.
Translocation Biosensors – Cellular System Integrators to Dissect CRM1-Dependent Nuclear Export by Chemicogenomics
2009
Fluorescent protein biosensors are powerful cellular systems biology tools for dissecting the complexity of cellular processes with high spatial and temporal resolution. As regulated nucleo-cytoplasmic transport is crucial for the modulation of numerous (patho)physiological cellular responses, a detailed understanding of its molecular mechanism would open up novel options for a rational manipulation of the cell. In contrast to genetic approaches, we here established and employed high-content cellular translocation biosensors applicable for dissecting nuclear export by chemicogenomics. A431 cell lines, stably expressing a translocation biosensor composed of glutathione S-transferase, GFP and…
Quantitation of antigen-reactive T cells in peripheral blood by IFNgamma-ELISPOT assay and chromium-release assay: a four-centre comparative trial
2000
The ELISPOT assay is increasingly being used for the monitoring of the induction of antigen-reactive T cells in cancer vaccination trials. In order to evaluate the reliability of T cell frequency analysis with the ELISPOT assay, a comparative study was performed in four European laboratories. Six samples from healthy subjects were analyzed for the frequency of influenza-reactive CD8+ T cells in peripheral blood mononuclear cells (PBMC) by IFNgamma-ELISPOT assay. In addition, one laboratory determined cytotoxic T cell precursor (CTL) frequencies in these samples by limiting dilution chromium-release assay (LDA), and three laboratories performed a variant of the LDA, the multiple microculture…
HIV-1 Adaptation to Antigen Processing Results in Population-Level Immune Evasion and Affects Subtype Diversification
2014
Summary The recent HIV-1 vaccine failures highlight the need to better understand virus-host interactions. One key question is why CD8+ T cell responses to two HIV-Gag regions are uniquely associated with delayed disease progression only in patients expressing a few rare HLA class I variants when these regions encode epitopes presented by ∼30 more common HLA variants. By combining epitope processing and computational analyses of the two HIV subtypes responsible for ∼60% of worldwide infections, we identified a hitherto unrecognized adaptation to the antigen-processing machinery through substitutions at subtype-specific motifs. Multiple HLA variants presenting epitopes situated next to a giv…
Alteration of DNA topoisomerase II activity during infection of H9 cells by human immunodeficiency virus type 1 in vitro: a target for potential ther…
1990
Infection of H9 cells with human immunodeficiency virus type 1 (HIV-1) was found to decrease the phosphorylation of DNA topoisomerase II during the initial phase of infection. Simultaneously, with a later overshoot of phosphorylation and the subsequent activation of DNA topoisomerase II, the production of HIV-1 started. Applying three new protein kinase C inhibitors from the class of O-alkylglycerophospholipids we demonstrated that inhibition of protein kinase C-mediated phosphorylation of DNA topoisomerase II resulted in an inhibition of HIV-1 production. Based on the differential effect of the two protein kinase C activators, phorbol ester and bryostatin, we conclude that phosphorylation …
“MIATA”—Minimal Information about T Cell Assays
2009
Immunotherapy, especially therapeutic vaccination, has a great deal of potential in the treatment of cancer and certain infectious diseases such as HIV (Allison et al., 2006; Fauci et al., 2008; Feldmann and Steinman, 2005). Numerous vaccine candidates have been tested in patients with a variety of tumor types and chronic viral diseases. Often, the best way to assess the clinical potential of these vaccines is to monitor the induced T cell response, and yet there are currently no standards for reporting these results. This letter is an effort to address this problem.
We study the important fi nding of a Merovingian denarius from Masilia (Marseille) in the Catalan city of Tarragona (Spain). Due to the scarcity of this coinage, not only in the Iberian Peninsula, we believe that its publicatio…
Gio Ponti : scena e design, un unico modo
2016
A 3D REWORKING OF THE URBAN TRANSFORMATIONS OF PALERMO IN RECENT HISTORY FOR A HYPOTHESIS OF A "CITY MUSEUM" BASED ON DIGITAL VISUALIZATIONS
2021
Abstract. The objective that drives this research is given by a multitude of information which, in addition to the contribution of technology, allows us to study, analyze, verify and remodel the sites, monuments and evolutions of the city through graphic processes of perspective restitution that start from the analysis of historical photos. The drawing methods, the digital graphic rendering and through the aid of geometric techniques, contribute to the reconstruction of projects and architectures that are now lost, this is possible thanks to the methods of perspective, axonometry and three-dimensional restitution. This remarkable photographic heritage belonging to Palermo, but also to any ot…
BEFORE THE EARTHQUAKE: RECONSTRUCTION AND PRESENTATION OF LOST SITES AND BUILDINGS
2021
Abstract. The paper presents the results of a research aiming at proving the efficacy of perspective restitution from photos for the reconstruction of buildings and sites that disappeared in the past century. The case study is the town of Messina, sited at the north-eastern corner of Sicily (Italy), facing the homonymous strait that divides Sicily from the Italian peninsula; in 1908 Messina and Reggio Calabria, at the opposite side of the strait, were levelled to the ground by a powerful earthquake, followed by a tsunami. No building or monument survived the destruction: a new town with the same name took the site of Messina. Except for few strongly reshaped buildings, the memory of the stre…
