Serum metabolome analysis by 1H-NMR reveals differences between chronic lymphocytic leukaemia molecular subgroups.
2010
Chronic lymphocytic leukaemia (CLL) is a heterogeneous disease exhibiting variable clinical course and survival rates. Mutational status of the immunoglobulin heavy chain variable regions (IGHVs) of CLL cells offers useful prognostic information for high-risk patients, but time and economical costs originally prevented it from being routinely used in a clinical setting. Instead, alternative markers of IGHV status, such as zeta-associated protein (ZAP70) or messenger RNA levels are often used. We report a (1)H-NMR-based metabolomics approach to examine serum metabolic profiles of early stage, untreated CLL patients (Binet stage A) classified on the basis of IGHV mutational status or ZAP70. M…
The cumulative amount of serum-free light chain is a strong prognosticator in chronic lymphocytic leukemia.
2011
AbstractIdentification of patients at risk of early disease progression is the mainstay of tailored management in chronic lymphocytic leukemia (CLL). Although application of established biomarkers is limited by intrinsic detection/readout complexities, abnormality of κ and λ serum-free light chain ratio [sFLC (κ/λ)] was proposed as a straightforward prognosticator in CLL. By analyzing 449 therapy-naive patients, we show that an abnormal sFLC(κ/λ), along with CD38, ZAP-70, IGHV mutations, cytogenetics and stage, independently predicts treatment-free survival (TFS) but becomes prognostically irrelevant if the cumulative amount of clonal and nonclonal FLCs [sFLC(κ + λ)], a variable associated …
Correlates of Human Herpesvirus-8 DNA detection among adults in Italy without Kaposi sarcoma.
2005
Background: The presence of Human Herpesvirus-8 (HHV8) DNA is predictive of Kaposi sarcoma (KS) among patients with HIV-associated or iatrogenic immunosuppression. However, correlates of HHV8-DNA detection in the general population remain undefined. Methods: We assessed correlates of HHV8-DNA detection among Italian adults without KS who had antibodies against HHV8-latent nuclear antigen by immunofluorescence assay. HHV8-K6 DNA sequences were detected in peripheral blood mononuclear cells using TaqMan CR. Results: Of the 158 subjects 26 (16.5%) had detectable HHV8-DNA [median copies/million cells, 53; (13-2128)]. Adjusted for age, sex, and laboratory, HHV8-DNA was detected more frequently i…
One size doesn’t fit all: a thematic analysis of interviews with people who have stopped participating in Narcotics Anonymous in Norway
2020
Abstract Background For persons with substance use disorders (SUDs), 12-step groups (TSGs) are the most available and used peer-based recovery resource, worldwide. However, disengagement is common, and attrition may partly be due to practices and procedures within these groups that are unacceptable to a portion of the population with SUDs. Our overall aim was to identify problematic issues related to Narcotics Anonymous (NA) participation in Norway, to inform addiction professionals’ strategies when referring persons to addiction-related self-help groups (SHGs). Methods In this qualitative study, we interviewed ten individuals who had previously participated regularly in NA for at least 6 m…
Identification of novel, clonally stable, somatic mutations targeting transcription factors PAX5 and NKX2-3, the epigenetic regulator LRIF1, and BRAF…
2021
Diagnosis of B-cell chronic lymphocytic leukemia (B-CLL) is usually straightforward, involving clinical, immunophenotypic (Matutes score), and (immuno)genetic analyses (to refine patient prognosis for treatment). CLL cases with atypical presentation (e.g., Matutes ≤ 3) are also encountered, and for these diseases, biology and prognostic impact are less clear. Here we report the genomic characterization of a case of atypical B-CLL in a 70-yr-old male patient; B-CLL cells showed a Matutes score of 3, chromosomal translocation t(14;18)(q32;q21) (BCL2/IGH), mutated IGHV, deletion 17p, and mutations in BCL2, NOTCH1 (subclonal), and TP53 (subclonal). Quite strikingly, a novel PAX5 mutation that w…
Structural study of gallium oxynitrides prepared by ammonolysis of different oxide precursors
2009
International audience; A comparative structural study has been carried out on gallium oxynitride powders using XRD and Raman spectroscopy. Gallium oxynitrides have been prepared by ammonolysis of either NiGa2O4 ternary oxide or the citrate method-derived amorphous oxide. Their crystal chemistry is different and appears to be influenced by the nature of the oxide precursor: whereas gallium oxynitride obtained from amorphous gallium oxide crystallizes with the common wurtzite structure, gallium oxynitride obtained from NiGa2O4 crystallizes with an original structure that we have identified as the carborundum II (B6) structure type or 6H-SiC. As far as we know, this is the first 6H-SiC struct…
Effect of Heat Exchange Transient Conditions with Moving Water-Air Interface on Space Charge Accumulation in Undersea HVdc Cables
2021
The accumulation of space charge inside the dielectric layer of an HVdc cable is one of the most important issue to be considered in the design stage and during the operating conditions. The separation of space charge is due to several factors including the dependence on temperature of the electrical conductivity of the insulation and the establishment of a thermal gradient under load conditions. This article is focused on the investigation of the effect of an axial heat transmission on the space charge and electric field distributions in an HVdc cable. In order to assess the impact of this phenomenon, a case study involving a cable immersed partly in water and partly in air has been simula…
Partial discharge behavior under single phase half and full-bridge rectifier
2017
The aim of the present work is to simulate partial discharges (PDs) phenomena under voltage stress close to the Direct Current (DC) waveform. The employed waveforms have been realized by using a full-wave and a half-wave rectifiers. After that, in order to generate a steadier voltage supply, a capacitive filter has been inserted in the output of the half-wave rectifier. Simulations have been carried out by modifying a previous software used in the case of Alternating Current (AC). Finally, experimental tests have been carried out in order to validate simulation results.
Testing the concentric-disk tension infiltrometer for field measurement of soil hydraulic conductivity
2010
Abstract The concentric-disk tension infiltrometer (CDTI) may be used to simultaneously measure the confined ( K c ) and the unconfined ( K u ) soil hydraulic conductivity, but it has received little testing. Comparison between K c and K u can be affected by the calculation approach applied to analyze unconfined steady-state infiltration rates. The objectives of this investigation were to: i) establish the effect of the calculation approach on the estimates of K u ; and ii) compare the K c and K u values measured by the CDTI. A clay soil, a structureless sandy loam soil, and a sandy loam soil with a relatively high gravel content were sampled using a disk of radius equal to 107.5 mm, compri…
