In an open clinical trial the authors treated 18 hospitalized patients suffering from endogenous depression with brofaromine (CGP 11305A), a competitive, selective, and short-acting inhibitor of type A monoamine oxidase (MAO). Four patients were defined as good responders, as they had a final HAMD score of between 0 and 7 points. Four patients were judged as improved, with final HAMD scores of between 8 and 15 points, while the remaining eight patients failed to respond (final HAMD score greater than or equal to 16 points). Abstract Background: Despite the long and widespread use of the Hamilton Depression Rating Scale (HAMD), standardized reliability studies in inexperienced raters are not available. Methods: Rater training was carried using three videotaped interviews with depressed patients in 21 psychiatric novices who had negligible previous experience with the HAMD. Chance-corrected coefficients of rating agreement with expert standards (weighted κ , ICC) were computed for single items and the total score of the HAMD. Ratings of the PANSS, HAMD, UKU, GAF, SOFAS, SWN-K and Kemp's compliance scale were performed and examined regarding their potential association with insight. Abstract The Hamilton Depression Scale (HAMD), Bech Rafaelsen Melancholia Scale (BRMS) and Montgomery Asberg Depression Rating Scale (MADRS) are analyzed according to mean discriminatory power, internal consistency, homogeneity and transferability. The analysis was done separately in different samples of patients with depressive syndromes: a) operationally defined depressive syndrome; b) Major Depressive Disorder (RDC); c) Major Depressive Disorder, endogenous type (RDC). BRMS and MADRS were superior to HAMD in all evaluated aspects. Further, the BRMS was superior to MADRS according to the criteria of homogeneity and transferability.

The effects of the optically active enantiomers of oxaprotiline (OXP), R(-) OXP and S(+) OXP, on depressive symptomatology and the sleep EEG were investigated in two separate exploratory studies. In addition, the neuroendocrine profile of both compounds was characterized in normal controls. In the patients treated with a daily oral dose of 150 mg S(+) OXP we found a Hamilton depression score that decreased from 29.1 +/- 1.8 (SEM) on day 0 to 14.7 +/- 3.2 on day 28 (P0.01). Abstract In Major Depressive Disorder, a growing data base suggests that the onset of antidepressants’ action can be detected by improvement of depressive symptoms in the first 10–14 days of treatment.
