Temperature‐Responsive Nanoparticles Enable Specific Binding of Apolipoproteins from Human Plasma

2021

Apolipoproteins are an important class of proteins because they provide a so-called stealth effect to nanoparticles. The stealth effect on nanocarriers leads to a reduced unspecific uptake into immune cells and thereby to a prolonged blood circulation time. Herein, a novel strategy to bind apolipoproteins specifically on nanoparticles by adjusting the temperature during their incubation in human plasma is presented. This specific binding, in turn, allows a control of the stealth behavior of the nanoparticles. Nanoparticles with a well-defined poly(N-isopropylacrylamide) shell are prepared, displaying a reversible change of hydrophobicity at a temperature around 32 °C. It is shown by label-f…

Oxygenated Cembrene Diterpenes from Sarcophyton convolutum: Cytotoxic Sarcoconvolutum A–E

2021

The soft coral genus Sarcophyton contains the enzymatic machinery to synthesize a multitude of cembrene-type diterpenes. Herein, highly oxygenated cembrenoids, sarcoconvolutum A–E (1–5) were purified and characterized from an ethyl acetate extract of the red sea soft coral, Sarcophyton convolutum. Compounds were assemblies according to spectroscopic methods including FTIR, 1D- and 2D-NMR as well as HRMS. Metabolite cytotoxicity was tested against lung adenocarcinoma, cervical cancer, and oral-cavity carcinoma (A549, HeLa and HSC-2, respectively). The most cytotoxic compound, (4) was observed to be active against cell lines A549 and HSC-2 with IC50 values of 49.70 and 53.17 μM, respectively.

Improved Efficacy of Synthesizing *MIII-Labeled DOTA Complexes in Binary Mixtures of Water and Organic Solvents. A Combined Radio- and Physicochemica…

2018

Typically, the synthesis of radiometal-based radiopharmaceuticals is performed in buffered aqueous solutions. We found that the presence of organic solvents like ethanol increased the radiolabeling yields of [68Ga]Ga-DOTA (DOTA = 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacatic acid). In the present study, the effect of organic cosolvents [ethanol (EtOH), isopropyl alcohol, and acetonitrile] on the radiolabeling yields of the macrocyclic chelator DOTA with several trivalent radiometals (gallium-68, scandium-44, and lutetium-177) was systematically investigated. Various binary water (H2O)/organic solvent mixtures allowed the radiolabeling of DOTA at a significantly lower temperature than …

New cyclic tetranuclear copper(II) complexes containing quadrilateral cores: Synthesis, structure, spectroscopy and their interactions with DNA in aq…

2017

Abstract Three new cyclic tetranuclear copper(II) complexes, Tetrakis{3-[(2-pyridylmethyl)-amino]-propionato}(tetrachloro)tetracopper(II)methanolhydrate (1·CH3OH·H2O), Tetrakis{3-[(2-pyridylmethyl)-amino]-propionato}(tetrathiocyanato)tetracopper(II) (2) and Tetrakis{3-[(2-pyridylmethyl)-amino]-propionato}(tetraazido)tetracopper(II) (3) have been synthesized by exploiting the chelating ability and bridging potential of a carboxyamine functionalized tridentate ligand, HL (HL = 3-[(2-Pyridylmethyl)-amino]-propionic acid). Complexes 1, 2 and 3 have been synthesized by carrying out reaction of the ligand HL with stoichiometric amounts of CuCl2·2H2O, CuCl2·2H2O/NH4SCN, and CuCl2·2H2O/NaN3, respec…

Halogen-containing BODIPY derivatives for photodynamic therapy

2019

Abstract Two iodinated (1 and 2) and two brominated (3 and 4) BODIPY-based photosensitizers are prepared and characterized using slightly modified synthetic procedures previously described. 1–4 present intense absorption bands in the 510–540 nm interval and very weak emissions due to the presence of halogen atoms in their structure. Irradiation of aqueous solutions of 1–4 with visible light (wavelength > 475 nm) induce the generation of singlet oxygen with quantum yields of ca. 0.62–0.66 for the photosensitizers containing iodine atoms (1 and 2) and of ca. 0.16–0.21 for the brominated counterparts (3 and 4). In vitro studies carried out with HeLa, SCC-13 and HaCaT cell lines and the four ph…

Attenuated total reflectance infrared determination of sodium nitrilotriacetate in alkaline liquid detergents

2005

Abstract An attenuated total reflectance infrared (ATR–FTIR) method has been developed for the direct determination of the chelating agent sodium nitrilotriacetate (NTA) in high pH cleaning formulations. Aqueous samples and standards were placed directly on the diamond ATR cell without any pre-treatment and FTIR spectra were recorded between 4000 and 600 cm−1. Measurement conditions were evaluated. Results obtained for five different commercial formulations containing from 6.22 to 9.25% (w/w) of NTA were in good agreement with the manufacturer's declared content (differences between 3.2 and −3.7%). Recovery studies evidenced the accuracy of the developed method, having found values between …

[3]Rotaxanes and [3]pseudorotaxanes with a rigid two-bidentate chelate axle threaded through two coordinating rings

2009

New [3]rotaxanes and [3]pseudorotaxanes have been synthesised using the “gathering and threading” effect of copper(I). By using click chemistry as the “stoppering” reaction, a good yield of the [3]rotaxane was obtained, either as a dicopper complex or as a metal-free compound after demetallation. The axle contains a central rigid aromatic block incorporating two bidentate chelates, and the threaded macrocycles are 30-membered rings. A model dicopper(I) [3]pseudorotaxane whose axle was end-functionalised by triisopropylsilyl groups could be crystallised and studied by X-ray diffraction. A particularly attractive structure was obtained showing a “slanted” geometry for the two rings and the ax…
