Selenium supplementation improves antioxidant capacity in vitro and in vivo in patients with coronary artery disease
2008
Background Selenium is a central determinant of antioxidative glutathione peroxidase 1 (GPx-1) expression and activity. The relevance of selenium supplementation on GPx-1 in coronary artery disease (CAD) needs to be established. We assessed the effect of selenium supplementation on GPx-1 in cell culture and on endothelial function in a prospective clinical trial. Methods Human coronary artery endothelial cells were incubated with 5.78 to 578 nmol/L sodium selenite, Se-methyl-selenocysteine hydrochloride, or seleno-l-methionine. Glutathione peroxidase 1 mRNA and protein expression and activity were measured. Coronary artery disease patients (n = 465) with impaired endothelial function (flow-…
Ischemia and post-ischemic regeneration of the small intestinal mucosa
1978
After ligation of the vascular arcades of the upper jejunum in rats, the ischemic damage to the intestinal mucosa and its regenerative behavior after ischemia lasting 120 minutes were investigated with histological and enzyme-histochemical methods. During the ischemic injury of the jejunal mucosa, there is rejection of hydropically swollen epithelial cells into the intestinal lumen advancing from the tip to the base of the villi without a previously detectable loss of activity of the enzymes investigated. At the end of ischemia lasting 120 minutes, there is complete destruction of the villi as well as the upper portions of the crypts. After rapid re-epithelialization of the mucosal surface …
Chapter 2 Single molecule spectroscopy of multichromophoric dendrimers at room and liquid helium temperatures
2004
Publisher Summary This chapter discusses the single molecule spectroscopy of multichromophoric dendrimers at room and liquid helium temperatures. Single molecule spectroscopy has become an important tool for the optical study of the electronic interactions of proximate chromophores as the electronic interactions can be studied without averaging over the properties of an intrinsically heterogeneous ensemble. Single perylenemonoimide (PMI) chromophores are selectively addressed in the frequency domain within spatially isolated multichromophoric dendrimers, which are dispersed in a polymer film. Although the chromophores are interacting, they still can be addressed separately. This feature in …
Reactive oxygen species mediate angiotensin II-induced leukocyte-endothelial cell interactions in vivo
2001
Abstract Chronically elevated angiotensin II (Ang-II)-induced hypertension is partly mediated by superoxide production. In this study, we have investigated whether the leukocyte-endothelial cell interactions elicited by Ang-II involve reactive oxygen species (ROS) generation. Intravital microscopy within the rat mesenteric microvessels was used. Superfusion (60 min) with Ang-II (1 nM) induced significant increases in leukocyte rolling flux, adhesion, and emigration, which were inhibited by pretreatment with superoxide dismutase or catalase. Dihydrorhodamine-123 oxidation indicated that ROS are primarily produced by the vessel wall. Administration of dimethylthiourea, desferrioxamine, or N-a…
Common antigenic determinant on ductular cells of normal pancreas, on mucosal cells of the gastrointestinal tract and on CEA
1990
CEA is widely used as a human tumour marker and was first defined by Gold and Freedman in 1965 as an antigenic component in cancers derived from gastrointestinal tract epithelium. It is a member of a large family of immunologically related glycoproteins that vary in size and tissue distribution. Studies with c-DNA clones for CEA and NCA reveal that this family consist only of a limited number of different proteins with variable glycosylation, due to post-transcriptional modifications. The complete gene family includes about 10 closely related genes. Recently it was published that in contrast to mRNA coding for CEA, mRNA coding for NCA was expressed predominantly in cancerous tissues. A mono…
Nitroglycerin-Induced Endothelial Dysfunction and Tolerance Involve Adverse Phosphorylation and S -Glutathionylation of Endothelial Nitric Oxide Synt…
2011
Objective— Continuous administration of nitroglycerin (GTN) causes tolerance and endothelial dysfunction by inducing reactive oxygen species (ROS) production from various enzymatic sources, such as mitochondria, NADPH oxidase, and an uncoupled endothelial nitric oxide synthase (eNOS). In the present study, we tested the effects of type 1 angiotensin (AT 1 )-receptor blockade with telmisartan on GTN-induced endothelial dysfunction in particular on eNOS phosphorylation and S -glutathionylation sites and the eNOS cofactor synthesizing enzyme GTP–cyclohydrolase I. Methods and Results— Wistar rats were treated with telmisartan (2.7 or 8 mg/kg per day PO for 10 days) and with GTN (50 mg/kg per d…

All-trans to 11-cis retinol isomerization in nuclear membrane fraction from bovine retinal pigment epithelium
1991
Abstract Isomerization of all-trans to 11-cis retinol has been studied in a membrane preparation from the nuclear fraction of bovine retinal pigment epithelium. When the nuclear membrane preparation deprived of endogenous retinoids is incubated with 4·5 μ m all-trans-retinol, the mean value calculated for the isomerase activity is 1·32 nmol 11-cis retinol formed hr−1 mg protein−1. Simultaneous formation of all-trans and 11-cis retinyl esters is also observed in the nuclear preparation. When assayed under the same experimental condition, RPE 150 000 g post-nuclear sediment shows about 70% of the isomerase activity found in the nuclear membrane fraction. Treatment of the nuclear membrane frac…
Trentino iedzīvotāju radzenes centrālās daļas endotēlijs
2017
