The histamine- or antihistamine — like activity of the resulting compounds was tested on isolated guinea pig ileum.

Theoretical study of selective H3 receptor antagonists of histamine

Abstract The conformations and charge distributions of three selective H3 receptor antagonists of histamine were determined using the MNDO approach. The results suggest that the conformational flexibilities of betahistine, N α-(2-phenylacetyl)histamine and thioperamide are different; however, the low-energy conformations of these compounds show closely related spatial orderings. The MNDO calculations predict a significant population of the N1H form in the imidazole systems of N α-(2-phenylacetyl)histamine and thioperamide. In vitro and in vivo clinical and experimental data have suggested that leukotrienes play a key role in inflammatory reactions of the skin. Antileukotriene drugs, i.e. leukotriene receptor antagonists and synthesis inhibitors, are a new class of anti-inflammatory drugs that have shown clinical efficacy in the management of asthma. Food allergy is a common and increasing problem worldwide. The newly-found knowledge might provide novel experimental strategies, especially for laboratory diagnosis. Approximately 20% of the population alters their diet for a perceived adverse reaction to food, but the application of double-blind placebo-controlled oral food challenge, the “gold standard” for diagnosis of food allergy, shows that questionnaire-based studies overestimate the prevalence of food allergies. Abstract Histamine has an important role in the process of the gastric mucosa inflammation acting via histamine receptor H2 (encoded by the gene HRH2). Single nucleotide polymorphism of the enhancer element of HRH2 gene promoter rs2067474 (1018G>A)may be associated with changes of expression of the receptor. We attempted to clarify the association of this polymorphism with gastric cancer and/or atrophic gastritis in the Latvian (Caucasian) population.
