Summary Protoplasts isolated from immature and mature mesophyll of wheat and oats were cultured in vitro . The potential progression from G1 (G0) to G2, through mitosis and cytokinesis was analysed. The degree and speed of progression through the cell cycle depended on the species and the state of maturity of the tissue from which the protoplasts were derived. In all the protoplast populations that were tested at least initiation of DNA synthesis was detected by labelling nuclei with the thymidine-analogue, bromodeoxyuridine (BrdU). Barley (Hordeum vulgare L.) leaves grow from the base and thus exhibit a smooth developmental gradient. Developing mesophyll cells acquire their typical lobed shape synchronously along this gradient. Successive changes in the patterns of cortical microtubules are involved in the shaping process. The changes include formation and dispersal of band-like structures, the establishment of a random network and a dramatic loss of microtubules after completion of cell shaping. The capacity of multi-layer perceptron artificial neural networks (MLP-ANN) and radial-basis function networks (RBFNs) to predict deoxynivalenol (DON) accumulation in barley seeds contaminated with Fusarium culmorum under different conditions has been assessed. Temperature (20-28 °C), water activity (0.94-0.98), inoculum size (7-15 mm diameter), and time were the inputs while DON concentration was the output. The dataset was used to train, validate and test many ANNs. Minimizing the mean-square error (MSE) was used to choose the optimal network. Composting organic residue is an interesting alternative to recycling waste as the compost obtained may be used as organic fertilizer. This study aims to assess the composting process of rice straw and sewage sludge on a pilot-scale, to evaluate both the quality of the composts obtained and the effects of applying such compost on soil properties and plant development in pot experiments. gamma-Tubulin is typically associated with microtubule organising centres, such as the centrosome, and appears to mediate microtubule nucleation. Centrosomes are usually not found in higher plants, but active genes homologous to gamma-tubulin have been identified in the plant kingdom, including the angiosperms Arabidopsis, maize and rice. We have isolated and characterised gamma-tubulin cDNA sequences of two further angiosperm species, barley and tobacco. In germinating grains of barley, Hordeum vulgare L. cv. Himalaya, free proline accumulated in the starchy endosperm during the period of rapid mobilization of reserve proteins. When starchy endosperms were separated from germinating grains and homogenized in a dilute buffer of pH 5 (the pH of the starchy endosperm), the liberation of proline continued in these suspensions. The process was completely inhibited by diisopropylfluorophosphate, indicating that it was totally dependent on serine carboxy-peptidases. Abstract Changes in the lipid composition of barley aleurone protoplasts following incubation in the presence or absence of gibberellic acid (GA) were studied and correlated with cell development. Analysis of neutral lipids in protoplast extracts and purified lipid bodies provide evidence for the strong effect of GA on the mobilization of storage lipids. In vivo protoplast staining with the membrane marker N -phenyl-l-naphthylamine (NPN) and phospholipid determinations of protoplast extracts show an increase in membrane lipids during protoplast incubation.
