A FRET-based random screening assay was used to generate hit compounds as sortase A inhibitors that allowed us to identify ethyl 3-oxo-2-(2-phenylhydrazinylidene)butanoate as an example of a new class of sortase A inhibitors. Other analogues were generated by changing the ethoxycarbonyl function for a carboxy, cyano or amide group, or introducing substituents in the phenyl ring of the ester and acid derivatives. The most active derivative found was 3-oxo-2-(2-(3,4dichlorophenyl)hydrazinylidene)butanoic acid (2b), showing an IC50 value of 50 µM. Single-cell analysis of population context advances RNAi screening at multiple levels

Isogenic cells in culture show strong variability, which arises from dynamic adaptations to the microenvironment of individual cells. Here we study the influence of the cell population context, which determines a single cell's microenvironment, in image‐based RNAi screens. We developed a comprehensive computational approach that employs Bayesian and multivariate methods at the single‐cell level. We applied these methods to 45 RNA interference screens of various sizes, including 7 druggable genome and 2 genome‐wide screens, analysing 17 different mammalian virus infections and four related cell physiological processes. Dipeptidyl Enoates As Potent Rhodesain Inhibitors That Display a Dual Mode of Action

Dipeptidyl enoates were prepared through a high-yielding two-step synthetic route. They have a dipeptidic structure with a 4-oxoenoate moiety as a warhead with multiple reactive sites. Dipeptidyl enoates were screened against rhodesain and human cathepsins B and L, and were found to be potent and selective inhibitors of rhodesain. Among them (S,E)-ethyl 5-((S)-2-{[(benzyloxy)carbonyl]amino}-3-phenylpropanamido)-7-methyl-4-oxooct-2-enoate (6) was the most potent, with an IC50 value of 16.4 nm and kinact/Ki=1.6×106 m−1 s−1 against rhodesain. Tutkimuksessa tarkasteltiin työn intensiivistymisen ilmenemismuotojen ja työn imun välisiä yhteyksiä yhdeksän ammattialan aineistossa (N = 7 786). Tutkimus toteutettiin kyselyllä, ja tuloksia analysoitiin regressioanalyyseillä ammattialoittain. Tulokset osoittivat, että yhteydet työn imuun vaihtelivat työn intensiivistymisen eri ilmenemismuotojen ja tason mukaan, mutta osin myös ammattialoittain. Kokemus työtahdin kiristymisestä oli yhteydessä heikompaan työn imuun lähes kaikilla ammattialoilla. Sen sijaan lisääntyneet oppimisvaatimukset työssä olivat joillakin ammattialoilla yhteydessä korkeampaan työn imuun. Toisaalta havaittiin, että työn imu on korkeinta silloin, kun oppimisvaatimukset …

Abstract Background This paper proposes a novel approach to the development of competence-oriented higher education, a national transformation aimed at harmonising and digitising undergraduate medical and dental education in Finland. Methods We apply phenomenography as a viable qualitative method for medical education research.
