Developmental and biochemical studies on the phenylalanine hydroxylation system in Drosophila melanogaster

Abstract The enzyme phenylalanine hydroxylase, the substrate phenylalanine, the product of the reaction tyrosine, and the probable in vivo cofactors (6R)- l -erytro-5,6,7,8-tetrahydrobiopterin (H4Bip) and 5,6,7,8-tetrahydropterin (H4Ptr), have been measured during development in Drosophila. The developmental profile of phenylalanine hydroxylase activity shows two peaks. The larger occurs at the time of pupation, coiciding with an important accumulation of tyrosine in the insect. The minor peak appears at the time of adult emergence. The developmental profile of H4Bip shows also two peaks, coinciding with those of maximal phenylalanine hydroxylase activity. Carotenoid-based integumental coloration is often associated with individual performance in various animals. This is because the limited amount of the pigment has to be allocated to different vital functions. However, most of the evidence for the carotenoid-based trade-off comes from vertebrate studies, and it is unclear if this principle can be applied to insects. Locust flight metabolism studied in vivo by 31P NMR spectroscopy

Flight metabolism of locusts has been extensively studied, but biochemical and physiological methods have led to conflicting results. For this reason the non-invasive and non-destructive method of 31P NMR spectroscopy was used to study migratory locusts, Locusta migratoria, at rest and during flight. 1. In the flight muscle of resting locusts the ratio of phosphoarginine to ATP was the same whether determined by NMR (1.76) or biochemically, but the NMR-visible content of inorganic phosphate (Pi) was only 40% of ATP, i.e., much lower than total Pi as determined biochemically. Two novel antimicrobial peptides, which we propose to name termicin and spinigerin, have been isolated from the fungus-growing termite Pseudacanthotermes spiniger (heterometabole insect, Isoptera). Termicin is a 36-amino acid residue antifungal peptide, with six cysteines arranged in a disulfide array similar to that of insect defensins. In contrast to most insect defensins, termicin is C-terminally amidated. Spinigerin consists of 25 amino acids and is devoid of cysteines. It is active against bacteria and fungi. Abstract The haemolymph of Spilostethus pandurus contains three female-specific proteins (I, II and III) as revealed by SDS-PAGE. Antibodies prepared against them recognize three fractions (4, 5 and 6) in ovarian extracts which have similar molecular weights suggesting that I, II and III are vitellogenins. Upon injection of a 14C amino acid mixture into vitellogenic females, labelled proteins first appear in the fat body, then in the haemolymph and next in the ovarian follicles.

Monitoring of arthropods focuses typically on changes in population and range size over time. Yet, there are a myriad of other aspects that could and should be monitored under the ongoing global and local environmental change. Stable isotope analysis, widely employed in short-term ecological studies, has potential in long-term monitoring of arthropods. Here we discuss the use of stable isotopes in monitoring terrestrial arthropods, provide some empirical examples of the use of bulk tissue samples in stable isotope analysis, and outline future directions in using compound-specific stable isotope analysis in monitoring. Dentro de un programa de conservación debe contemplarse este riesgo como otro más a evaluar, tomar medidas para su prevención y, en último término, actuar con métodos de erradicación en caso de que la situación lo requiera. El Control Integral de Plagas (del inglés Integrated Pest Management -IPM-) aplicado por primera vez en la agricultura ecológica para disminuir el uso de pesticidas, es la herramienta fundamental para gestionar este factor de deterioro.
