A human hepatocellular in vitro model to investigate steatosis

The present study was designed to define an experimental model of hepatocellular steatosis with a fat overaccumulation profile in which the metabolic and cytotoxic/apoptotic effects could be separated. This was accomplished by defining the experimental conditions of lipid exposure that lead to significant intracellular fat accumulation in the absence of overt cytotoxicity, therefore allowing to differentiate between cytotoxic and apoptotic effects. By immunolabeling of cryosections, we have characterized in rat cardiac myocytes the cation-independent mannose-6-phosphate receptor (MPR), a lysosomal membrane glycoprotein, lgp120, and a lysosomal enzyme, MEP (homologous to cathepsin L). Most of the MPR label was located in large membrane-filled structures (MPR structures) in large clusters of mitochondria adjacent to but distinct from the Golgi complex. Lpg120 and MEP showed typical lysosomal localization throughout the cell, often associated with regions that appeared to contain autophagosome-like structures. The melatonin metabolism in the mammalian pineal gland is under the clear influence of sympathetic fibers originating in the superior cervical ganglia (SCG). Previous studies suggested that also pineal ‘synaptic’ ribbons (SR) are regulated by the gland’s sympathetic innervation. To gain more insight into the mechanisms involved SR numbers were determined in adult rats which were chemically sympathectomized as newborns by injection of 6-hydroxydopamine (6-OHDA). In control animals (saline injections), a clear day/night difference in the number of SR is present. Acidosis and in particular lactacidosis from anaerobic metabolism are considered of primary significance among the consequences of cerebral ischemia, seizures,, and head injury (Siesjo 1981). A marked decrease in brain tissue pH has been demonstrated in cerebral ischemia - even more so in hyperglycemic subjects - where lactic acid may accumulate to 20–30 mM and higher concentrations (Rehncrona et al. 1980; Katsura et al. 1991). Tissue pH may drop to pH 5.5 (Chopp et al. 1988). In consequence, acidosis has since long been suspected as a mediator of brain damage (Siesjo 1981).

This study examined the intrarater and interrater reliability of the Test of Gross Motor Development—3rd Edition (TGMD-3). Participants were 60 Finnish children aged between 3 and 9 years, divided into three separate samples of 20. Two samples of 20 were used to examine the intrarater reliability of two different assessors, and the third sample of 20 was used to establish interrater reliability. Children’s TGMD-3 performances were video-recorded and later assessed using an intraclass correlation coefficient, a kappa statistic, and a percent agreement calculation. Aim The World Health Organization Disability Assessment Schedule II (WHODAS II) was developed for assessing disability. This study provides data on the validity and utility of the Spanish version of the WHODAS II in a large sample of patients with schizophrenia. Methods The sample included 352 patients with a schizophrenia spectrum disorder.
