Characterization of 150 Wheat Cultivars by LC-MS-Based Label-Free Quantitative Proteomics Unravels Possibilities to Design Wheat Better for Baking Qu…

2021

Wheat (Triticum aestivum ssp. aestivum) contributes to 20% of the human protein supply, delivers essential amino acids and is of fundamental importance for bread and pasta quality. Wheat proteins are also involved in adverse human reactions like celiac disease (CD), wheat allergy (WA) and non-celiac wheat sensitivity (NCWS). Using liquid chromatography-mass spectrometry (LC-MS)-based label-free quantitative (LFQ) proteomics of aqueous flour extracts, we determined 756 proteins across 150 wheat cultivars grown in three environments. However, only 303 proteins were stably expressed across all environments in at least one cultivar and only 89 proteins thereof across all 150 cultivars. This und…

Terpenoid bio-transformations and applications via cell/organ cultures: a systematic review.

2019

Structurally diverse natural products are valued for their targeted biological activity. The challenge of working with such metabolites is their low natural abundance and complex structure, often with multiple stereocenters, precludes large-scale or unsophisticated chemical synthesis. Since select plants contain the enzymatic machinery necessary to produce specialized compounds, tissue cultures can be used to achieve key transformations for large-scale chemical and/or pharmaceutical applications. In this context, plant tissue-culture bio-transformations have demonstrated great promise in the preparation of pharmaceutical products. This review describes the capacity of cultured plant cells t…

Anthropologie physiologique

2007

Monitoring of transglutaminase crosslinking reaction by 1H NMR spectroscopy on model substrates

2015

International audience; A new method based on 1H NMR spectroscopy was developed for monitoring transglutaminase crosslinking reaction with model molecules (CBZ-Gln-Gly and N-α-acetyl-lysine). The transglutaminase reaction led to the appearance of new resonances on NMR spectrum as well as significant decrease in others. The new observed resonances, originated from newly formed ɛ-(γ-glutamyl)lysine isopeptide bonds, evidence the enzymatic reaction and allow to quantify the ɛ-(γ-glutamyl)lysine fragment. Moreover, the decrease in resonance intensity, originated from lysine, permit to determine the crosslinking degree. These results obtained by 1H NMR spectroscopy can be used as an alternative …

Phylogenomics Identifies an Ancestral Burst of Gene Duplications Predating the Diversification of Aphidomorpha

2019

Aphids (Aphidoidea) are a diverse group of hemipteran insects that feed on plant phloem sap. A common finding in studies of aphid genomes is the presence of a large number of duplicated genes. However, when these duplications occurred remains unclear, partly due to the high relatedness of sequenced species. To better understand the origin of aphid duplications we sequenced and assembled the genome of Cinara cedri, an early branching lineage (Lachninae) of the Aphididae family. We performed a phylogenomic comparison of this genome with 20 other sequenced genomes, including the available genomes of five other aphids, along with the transcriptomes of two species belonging to Adelgidae (a close…

Immunity and other defenses in pea aphids, Acyrthosiphon pisum

2010

Background Recent genomic analyses of arthropod defense mechanisms suggest conservation of key elements underlying responses to pathogens, parasites and stresses. At the center of pathogen-induced immune responses are signaling pathways triggered by the recognition of fungal, bacterial and viral signatures. These pathways result in the production of response molecules, such as antimicrobial peptides and lysozymes, which degrade or destroy invaders. Using the recently sequenced genome of the pea aphid (Acyrthosiphon pisum), we conducted the first extensive annotation of the immune and stress gene repertoire of a hemipterous insect, which is phylogenetically distantly related to previously ch…

Anti-Bacterial Activity and Online HPLC-DPPH Based Antiradical Kinetics of Medicinal Plant Extracts of High Relevance for Cosmetics Production

2018

The present study provides an assessment of extracts of six medicinal plants for their applicability in cosmetic formulations aimed at anti-UV protection and anti-acne treatment. Ethanol extracts of Chelidonium majus L., Myrtus communis L., Hamamelis virginiana L., Juniperus communis L., Alchemilla vulgaris and Ilex paraguariensis are analysed for the phytochemical content, antimicrobial activity, antiradical activity and antiradical kinetics. The phytochemical composition of the plants is investigated by LC-MS analysis and the most active antioxidants in plant extracts are determined by online HPLC-DPPH assay. The kinetic parameters of the antiradical reaction are determined both for the s…

Improvement of resveratrol production from waste residue of grape seed by biotransformation of edible immobilized Aspergillus oryzae cells and negati…

2017

Abstract A biotransformation method of producing resveratrol with immobilized Aspergillus oryzae negative pressure cavitation bioreactor (ICNPCB) with biphasic ionic liquid aqueous system pretreatment was investigated. Subsequently, the biotransformation conditions were optimized for the resveratrol production under types of ILs [C4MIM]Br, concentration of ILs 0.75 M, temperature 31 °C, ratios of solid to liquid 1:15 (mg/L), time 60 h and pH 6.0, the yield of resveratrol reached 224.61 ± 0.35 μg/g, which increased 4.36-fold to that of untreated one. The conversion rate of piceid reached 85.21%. The residual activity of immobilized microorganism was 82.26% after used for 15 runs. The develop…
