Monitoring of transglutaminase crosslinking reaction by 1H NMR spectroscopy on model substrates

International audience; A new method based on 1H NMR spectroscopy was developed for monitoring transglutaminase crosslinking reaction with model molecules (CBZ-Gln-Gly and N-α-acetyl-lysine). The transglutaminase reaction led to the appearance of new resonances on NMR spectrum as well as significant decrease in others. The new observed resonances, originated from newly formed ɛ-(γ-glutamyl)lysine isopeptide bonds, evidence the enzymatic reaction and allow to quantify the ɛ-(γ-glutamyl)lysine fragment. Impacts of chitinase-transformed silver birch on leaf decomposition and soil organisms

Genetically manipulated silver birch (Betula pendula) leaves were used in microcosms experiments to evaluate their impacts on different groups of decomposing soil fauna. Birches were transformed to produce chitinase IV from sugar beet. We compared decomposition rates of leaves, growth and reproduction of soil fauna deriving nutrition from these leaves. Population numbers of collembolans (Folsomia candida and Lepidocyrtus lignorum) and nematodes were measured and decomposition rates of the birch leaves were recorded. ABSTRACT With the aim of obtaining new strategies to control plant diseases, we investigated the ability of antagonistic lipopolypeptides (paenimyxin) from Paenibacillus sp. strain B2 to elicit hydrogen peroxide (H 2 O 2 ) production and several defense-related genes in the model legume Medicago truncatula . For this purpose, M. truncatula cell suspensions were used and a pathosystem between M. truncatula and Fusarium acuminatum was established. Arginase induction can play a defensive role through the reduction of arginine availability for phytophageous insects. Arginase activity is also induced during gall growth caused by Plasmodiophora brassicae infection in roots of Arabidopsis thaliana; however, its possible role in this context has been unclear. We report here that the mutation of the arginase-encoding gene ARGAH2 abrogates clubroot-induced arginase activity and results in enhanced gall size in infected roots, suggesting that arginase plays a defensive role.
