Inflammasome activation in Ankylosing Spondylitis is associated to gut dysbiosis

Objective: We undertook this study to evaluate the activation and functional relevance of inflammasome pathways in ankylosing spondylitis (AS) patients and rodent models and their relationship to dysbiosis. Methods: An inflammasome pathway was evaluated in the gut and peripheral blood from 40 AS patients using quantitative reverse transcriptase–polymerase chain reaction (qRT-PCR), immunohistochemistry (IHC), flow cytometry, and confocal microscopy, and was compared to that of 20 healthy controls and 10 patients with Crohn’s disease. Abstract Borylation of lithium indenide with BBrMe 2 gives the allylic 1-indenyl derivative (1-Ind)BMe 2 ( 1c ) together with the bis-allylic connected species (1-Ind) 2 BMe ( 3 ) and with BCl i Pr 2 , the allylic 1-indenyl (1-Ind)B i Pr 2 ( 1d ); the latter, with Et 3 N or Py added, rearranges to the vinylic 3-indenyl isomer (3-Ind)B i Pr 2 ( 2d ). The disubstituted indene C 9 H 6 Ind(SiMe 3 )B(O 2 C 2 Me 4 ) ( 5 ) was prepared from Li[C 9 H 6 B(O 2 C 2 Me 4 )] ( 4b ) and Me 3 SiCl. Abstract The reaction of ortho-lithiated N,N-dimethyl-1-arylethyl-amines with triphenyl- boron leads, with elimination of phenyllithium, to (R)-(−)-2,2,3-trimethyl-1,1-diphenyl -1H-2-azonia-1-borata-indene (1), 2,3-dihydro-1,1,2-trimethyl-3,3-diphenyl- 1H-2-azonia-3-borata-benz[e]indene (2) and 2,3-dihydro-2,2,3-trimethyl-1,1-diphenyl -1H-2-azonia-1-borata-benz[e]indene (3) with a strongB←N bond. 1, 2 and 3 are stable against oxygen, water and alcohols. Triarylboron compounds sensitive to the same reagents are formed by the reaction of triphenylboron with some aryllithium compounds substituted with electron-donating groups.

PEG-benzofulvene copolymer hydrogels for antibody delivery.

A new amphiphilic copolymer have been synthesized starting from the hydrosoluble polyaspartylhydrazide (PAHy) polymer, by grafting both hydrophilic PEG(2000) chains and hydrophobic palmitic acid (C(16)) moieties on polymer backbone, and the structure of obtained PAHy-PEG(2000)-C(16) copolymer have been characterized by 2D (1)H/(13)C NMR experiments. PAHy-PEG(2000)-C(16) copolymer showed the ability of self-assembling in aqueous media giving a core-shell structure and resulted potentially useful for encapsulating and dissolving hydrophobic drug. Characterization of antihistamine–human serum protein interactions by capillary electrophoresis

An important topic in the drug discovery and development process is the role of drug binding to plasma proteins. In this paper the characterization of the interaction between antihistamines (cationic drugs) towards human serum albumin (HSA) and alpha(1)-acid glycoprotein (AGP) under physiological conditions by capillary electrophoresis-frontal analysis is presented. Furthermore, the binding of these drugs to all plasma proteins is evaluated by using ultrafiltration and capillary electrophoresis. A theoretical study of the emission spectra of indole and its analogs: indene, benzimidazole, and 7-azaindole

Abstract The complete active space (CAS) SCF method and multiconfigurational second-order perturbation theory (CASPT2) have been used to study the electronic spectra of indole, indene, benzimidazole, and 7-azaindole. The paper is focused on the study of the low-lying valence triplet and singlet electronic states at the optimized geometries of the excited states. The geometries have been optimized by using analytic CASSCF derivatives. CASPT2 point calculations have been performed in order to obtain band origins and relaxed emission energies. Mittels 14C- und 2H-indizierten Verbindungen werden die reversible, nucleophile Addition von Benzol an Zimtaldehyd sowie die 1.3-Wasserstoff-Verschiebung im Crotonaldehyd beim Ringschlus nachgewiesen. On the Mechanism of the Indene Formation by Friedel-Crafts Cycloalkenylation α,β-Unsaturated aldehydes were condensed with benzene in a Friedel-Crafts type reaction to give indenes. With the aid of 14C- and 2H-labelled compounds both the reversible nucleophilic addition of benzene to cinnamaldehyde and the 1.3-hydrogen shift in the cycloalkenylation of crotonaldehyde are examined.

Dimerization of the kinase ARAF promotes MAPK pathway activation and cell migration.

The RAF family of kinases mediates RAS signaling, and RAF inhibitors can be effective for treating tumors with BRAF V600E mutant protein. However, RAF inhibitors paradoxically accelerate metastasis in RAS -mutant tumors and become ineffective in BRAF V600E tumors because of reactivation of downstream mitogen-activated protein kinase (MAPK) signaling. We found that the RAF isoform ARAF has an obligatory role in promoting MAPK activity and cell migration in a cell type–dependent manner. Evaluation of thermoresponsive properties and biocompatibility of polybenzofulvene aggregates for leuprolide delivery

Abstract In this study, a polybenzofulvene derivative named poly-6-MOEG-9-BF3k, was evaluated as polymeric material for the production of injectable thermoresponsive nano-aggregates able to load low molecular weight peptidic drug, like the anticancer leuprolide. Thermoresponsive behavior of poly-6-MOEG-9-BF3k was studied in aqueous media by evaluating scattering intensity variations by means of DLS in function of temperature.
