The fischer indole synthesis of 8-methyl-5-substituted-1-oxo-β-carbolines: A remarkable high yield of a [1,2]-methyl migration.
1991
Abstract A very unusual high yield of rearranged products (via methyl migration) has been obtained in the Fischer indole cyclization of the 3-(2,5-dimethyl phenylhydrazone) of 2,3-piperidine dione (6b). When a less activated aromatic ring is present in the phenylhydrazone 6, such a behaviour is not observed and cyclization occurs essentially without rearrangement.
ChemInform Abstract: First Electrophilic Substitutions of 3-Substituted Indoles with Diethoxycarbenium Tetrafluoroborate: Functionalized Indole Deriv…
1990
The indoles 2a-2c react with diethoxycarbenium tetrafluoroborate (1) to furnish the indolecarbaldehydes 3a-3d. In the thermodynamically controlled reaction of 3-methylindole (2a) with 1 the tris(indolyl)methane 4 and diskatole (5), are formed in addition. The limitations of these reactions are discussed and evidence is presented for a C-3-ipso-attack and a Wagner-Meerwein rearrangement, respectively, leading to the formation of 3b or 3d. A theoretical study of the emission spectra of indole and its analogs: indene, benzimidazole, and 7-azaindole
2000
Abstract The complete active space (CAS) SCF method and multiconfigurational second-order perturbation theory (CASPT2) have been used to study the electronic spectra of indole, indene, benzimidazole, and 7-azaindole. The paper is focused on the study of the low-lying valence triplet and singlet electronic states at the optimized geometries of the excited states. The geometries have been optimized by using analytic CASSCF derivatives. CASPT2 point calculations have been performed in order to obtain band origins and relaxed emission energies. The results are analyzed in the context of the complex emission processes, both fluorescence and phosphorescence, displayed by the title compounds, whic…
5′-Benzylidene-1′′-methyl-4′′-phenyltrispiro[1,3-dioxolane-2,1′-cyclohexane-3′,3′′-pyrrolidine-2′′,3′′′-indole]-4′,2′′′-dione
2017
In the title compound, C32H30N2O4, two spiro links connect the methyl-substituted pyrrolidine ring to the oxindole and cyclohexanone rings. The cyclohexanone ring is further connected to the dioxalane ring by a third spiro junction. Both the pyrrolidine and dioxalane rings adopt a twist conformation. The indole ring is nearly planar, with a maximum deviation of 0.0296 (7) Å, and the cyclohexanone ring adopts a distorted boat conformation. In the crystal, C—H...O and N—H...N hydrogen-bonding interactions connect molecules into chains running parallel to thebaxis, which are further linked into layers parallel to theabplane by C—H...O hydrogen bonds.
Essential on the Photophysics and Photochemistry of the Indole Chromophore by Using a Totally Unconstrained Theoretical Approach
2015
Indole is a chromophore present in many different molecules of biological interest, such as the essential amino acid tryptophan and the neurotransmitter serotonin. On the basis of CASPT2//CASSCF quantum chemical calculations, the photophysical properties of the system after UV irradiation have been studied through the exploration of the potential energy hypersurfaces of the singlet and triplet low-lying valence excited states. In contrast to previous studies, the present work has been carried out without imposing any restriction to the geometry of the molecule (C1 symmetry) and by performing minimum energy path calculations, which is the only instrument able to provide the lowest-energy evo…
Induction of alkaloid diversity in hybrid plant cell cultures
1999
The treatment of Rauwolfia serpentina x Rhazya stricta somatic hybrid cell suspension culture with 100 μM of methyl jasmonate led to a general increase in indole alkaloid content and to qualitative changes in the alkaloid pattern. The content of Six alkaloids were investigated with respect to their content in both the cell biomass and nutrition medium. Intracellular 17-O-acetyl-norajmaline content on the 5th day after treatment had increased about 40-fold compared with the control culture. The respective concentrations of the other alkaloids increased by a factor of two to five. In total 26 indole alkaloids were identified in extracts of the methyl jasmonate-treated culture by TLC, UV, MS a…
Diels-Alder reaction between indoles and cyclohexadienes photocatalyzed by a (thia)pyrylium salt
2007
"Thiapyrylium salt 1 is an efficient photocatalyst in the Diels-Alder reaction between indoles (InHs) and 1,3-cyclohexadienes (CHDs). For instance, irradiation of deaerated dichloromethane solutions containing 1 (1 mM), indole (20 mM) and cyclohexadiene (40 mM), in the presence of an acylating agent, led to the Diels-Alder cycloadduct in 62% yield (endo:exo ratio of 1.8:1). Taking into account the very high intersystem crossing yield (Φisc = 0.97) of thiapyrylium salt 1, involvement of its triplet excited state in this Diels-Alder reaction has been studied using steadystate and time-resolved experiments; the results are compared with those previously obtained for pyrylium salts, which exhib…
1,2,4-Oxadiazole topsentin analogs as staphylococcal biofilm inhibitors targeting the bacterial transpeptidase sortase A
2020
The inhibition or prevention of biofilm formation represents an emerging strategy in the war against antibiotic resistance, interfering with key players in bacterial virulence. This approach includes the inhibition of the catalytic activity of transpeptidase sortase A (Srt A), a membrane enzyme responsible for covalently attaching a wide variety of adhesive matrix molecules to the peptidoglycan cell wall in Gram-positive strains. A new series of seventeen 1,2,4-oxadiazole derivatives was efficiently synthesized and screened as potential new anti-virulence agents. The ability of inhibiting biofilm formation was evaluated against both Gram-positive and Gram-negative pathogens. Remarkably, all…
2,6-Disubstituted imidazo[2,1-b][1,3,4]thiadiazole derivatives as potent staphylococcal biofilm inhibitors.
2019
Abstract A class of 36 new 2-(6-phenylimidazo[2,-1-b][1,3,4]thiadiazol-2-yl)-1H-indoles was efficiently synthesized and evaluated for their anti-biofilm properties against the Gram-positive bacterial reference strains Staphylococcus aureus ATCC 25923, S. aureus ATCC 6538 and Staphylococcus epidermidis ATCC 12228, and the Gram-negative strains Pseudomonas aeruginosa ATCC 15442 and Escherichia coli ATCC 25922. Many of these new compounds, were able to inhibit biofilm formation of the tested staphylococcal strains showing BIC50 lower than 10 μg/ml. In particular, derivatives 9c and 9h showed remarkable anti-biofilm activity against S. aureus ATCC 25923 with BIC50 values of 0.5 and 0.8 μg/ml, r…
Inhibition of astroglial cell proliferation by alcohols: interference with the protein kinase C-phospholipase D signaling pathway.
2000
Abstract Ethanol inhibits astroglial cell proliferation, an effect that may contribute to the development of alcoholic embryopathy in humans. In the present study, we investigated inhibitory effects of ethanol and butanol isomers (1-, 2- and t -butanol) on astroglial cell proliferation induced by the strongly mitogenic phorbol ester, 4s-phorbol-12α,13s-dibutyrate (PDB). 4s-Phorbol-12α,13s-dibutyrate (PDB) induced a 10-fold increase of [3H]thymidine incorporation in cortical astrocytes prepared from newborn rats (EC 50 : 70 nM) which was blocked by Ro 31-8220, a cell-permeable protein kinase C (PKC) inhibitor. Ethanol blocked PDB-induced astroglial proliferation in a concentration-dependent …
