The influence of alginate-embedding on the maintenance of functioning and morphological integrity in long-term culture of isolated porcine islets and islet cells was studied. Free-floating islets and islet cells served for control. Function was tested after the 1st, 2nd and 4th week. Basal and glucose-stimulated insulin secretion of embedded islets decreased slightly, but significantly after the first week (from 4.39 +/- 0.64 to 2.87 +/- 0.47 at normal and from 11.96 +/- 1.44 to 4.76 +/- 0.78 microU pro 24 h pro islet at elevated glucose concentration, p < 0.05 and < 0.01, resp.) and remained unchanged thereafter. Ghrelin is a peptide hormone produced mainly in the stomach that has widespread tissue distribution and diverse hormonal, metabolic and cardiovascular activities. The circulating ghrelin concentration increases during fasting and decreases after food intake. Ghrelin secretion may thus be initiated by food intake and is possibly controlled by nutritional factors. Lean subjects have increased levels of circulating ghrelin compared with obese subjects. The 11 C-labeled sulfonylurea receptor 1 (SUR1) ligand (S)-2-(( 11 C)methoxy)-4-(3-methyl-1-(2-piperidine-1-yl-phenyl)- butyl-carbamoyl)-benzoic acid (( 11 C)methoxy-repaglinide) was synthesized in an overall radiochemical yield of 35% after 55 min with a radiochemical purity higher than 99%. Risposta insulo-secretrice al carico endovenoso di tolbutamide

Gli AA. hanno studiato il comportamento della risposta insulinemica dopo stimolazione con tolbutamide endovenosa in un piccolo gruppo di soggetti normali normosomici, obesi non diabetici e diabetici obesi. In tutti i soggetti il massimo aumento dell'insulinemia (determinata con metodo radio-immunologico) compare a 5 mp ed e maggiore negli obesi rispetto ai normali e ai diabetici. La caduta percentuale del picco insulinemico dimostra un andamento parallelo nei normali e negli obesi, mentre e molto piu lenta nei diabetici obesi. Vengono discusse le cause di questo comportamento e la natura dell'iperinsulinismo presente nell'obesita con e senza diminuita tolleranza agli idrati di carbonio.
