Chromatographic Efficiency in Micellar Liquid Chromatography: Should it Be Still a Topic of Concern?

Micellar liquid chromatography (MLC) was first proposed as an attractive alternative to avoid the use of organic solvents. It was soon apparent that pure micellar solutions yield poor efficiencies. This problem was remediated by the addition of a small amount of an organic solvent. However, the general opinion of the poor peak shape has prevailed as a handicap for MLC, in spite of the fact that the hybrid mode often offers similar or even improved efficiencies (for basic compounds) relative to that attained in the hydro-organic mode. A simple and flexible system, employing a column switching technique, has been designed to allow the analysis of peptides and proteins smaller than 15 kDa by molecular weight in filtered urine samples by performing a direct on-column injection utilising simultaneous sample clean-up and trace enrichment. The positively charged peptides and small proteins in the sample are attracted to the inner, negatively charged pore structure of the RAM-SCX column while the larger proteins and uncharged or negatively charged compounds are excluded. Gel permeation Chromatography in coiled columns

Whereas in gas chromatography the use of coiled columns is the usual procedure, in liquid chromatography the use of straight columns is favoured. Bent columns give an additional contribution to peak spreading which is of greater importance in liquid chromatography than in gas chromatography owing to smaller diffusion coefficients. The low linear velocities of the mobile phase used in gel permeation chromatography enable conditions to be found where the additional contribution of bending to peak spreading is small. Purification and identification of a cytokinin from moss callus cells.

A cytokinin was isolated from the culture medium of callus cells of the moss hybridFunaria hygrometrica (L.) Sibth xPhyscomitrium piriforme Brid. The purification procedure included ethyl-acetate extraction, silver-salt precipitation, crystallization as picrate, and ion exchange chromatography. The structure of the cytokinin was confirmed as N(6)-(Δ(2)-isopentenyl)adenine by means of gas chromatography and mass spectrometry. The concentration of the compound in the culture medium was determined at ca. 10(-6) M.

Thin-layer chromatography of 2-methyl-4-chlorophenoxyacetic acid and its soil metabolites

Abstract The thin-layer chromatography of 2-methyl-4-chlorophenoxyacetic acid, 4-chloro- o -cresol and 3-methyl-5-chlorocatechol and their pentafluorobenzyl derivatives has been studied on silica gel as adsorbent with 19 solvent systems. The best separation of the individual components occurred with toluene-benzene-acetic acid (2:2:1). Chloroform-diethyl ether-toluene (1:1:1) was suitable for the group separation of the pentafluorobenzyl derivatives.

Size Exclusion Chromatography for Semipreparative Scale Separation of Au 38 (SR) 24 and Au 40 (SR) 24 and Larger Clusters

Size exclusion chromatography (SEC) on a semipreparative scale (10 mg and more) was used to size-select ultrasmall gold nanoclusters (2 nm) from polydisperse mixtures. In particular, the ubiquitous byproducts of the etching process toward Au(38)(SR)(24) (SR, thiolate) clusters were separated and gained in high monodispersity (based on mass spectrometry). The isolated fractions were characterized by UV-vis spectroscopy, MALDI mass spectrometry, HPLC, and electron microscopy. Most notably, the separation of Au(38)(SR)(24) and Au(40)(SR)(24) clusters is demonstrated.

Porous fractal gels: secondary effects in SEC

Abstract The fractal nature of porous silica gels, Spherosil XOA 200 and of a Lichrospher mixture, has been tested through size exclusion chromatography of polystyrene in pure and mixed eluents. Fractal parameters, D f and L , depend on eluent with D f values close to 2.0 for Spherosil and higher than 2.0 for Lichrospher. D f and L changes with eluent have been related to secondary separation effects and partition and adsorption contributions have been evaluated.

Accurate determination of element species by on-line coupling of chromatographic systems with ICP-MS using isotope dilution technique

Abstract The instrumental design for coupling different liquid chromatographic systems such as ion, reversed phase, and size exclusion chromatography as well as capillary gas chromatography, with ICP-MS for the determination of element species is described. Estimation of the Compatibility Between Poly(Methylmethacrylate) and Poly(Styrene Co Vinyl Phenol) Blends from Dilute Solution Measurements

Abstract The compatibility of poly(methyl methacrylate) (PMMA) with poly(styrene‐co‐vinyl phenol) (PS‐VPh) with two different contents of vinyl phenol, 5.8 and 7.2%, in dilute tetrahydrofuran solutions has been investigated by size exclusion chromatography and fluorescence spectroscopy at 25°C. The chromatographic technique permits the evaluation of the preferential solvation at different PMMA/PS‐VPh ratios. Changes in the fluorescence properties of PS‐VPh, caused by its association with PMMA, were used to obtain the fraction of copolymer bound to PMMA at diverse PMMA compositions.
