Dispersion management in two-photon microscopy by using diffractive optical elements.

We demonstrate efficient generation of wide-field fluorescence signals in two-photon microscopy exploiting diffractive optical elements and short pulses by using a dispersion-compensated beam delivery optics module. Computer-generated holograms are codified onto a phase-only spatial light modulator, which allows for arbitrary single-shot patterning of the sample. Spatiotemporal shaping of the pulse is mandatory to overcome spatial chirp and pulse-front tilt effects that spread both in space and time the irradiance patterns, thus limiting not only the spatial resolution but also the signal-to-noise ratio in two-photon microscopy. How and why does willow biochar increase a clay soil water retention capacity?

Addition of biochar into a soil changes its water retention properties by modifying soil textural and structural properties. In addition, internal micrometer-scale porosity that is able to directly store readily plant available water affects soil water retention properties. This study shows how precise knowledge of the internal micrometer-scale pore size distribution of biochar can deepen the understanding of the biochar-water interactions in soils. Evaluation of the use of wavefront encoding to reduce depth-induced aberration in structured-illumination microscopy

Three-dimensional imaging is affected by depth-induced spherical aberration (SA) when imaging deep into an optically thick sample. In this work, we evaluate the impact of SA on the performance of incoherent grating-projection structured illumination microscopy (SIM). In particular, we analyze the reduction of the contrast in the structured pattern and compare the reconstructed SIM images for different amounts of SA. In order to mitigate the impact of SA, we implement and evaluate in SIM a wavefront encoded imaging system using a square cubic (SQUBIC) phase mask, an approach shown previously to be successful in conventional microscopy.
Leakage radiation microscopy of surface plasmon coupled emission: investigation of gain-assisted propagation in an integrated plasmonic waveguide.

International audience; Using a single-mode dielectric-loaded surface plasmon polariton waveguide doped with quantum dots, we were able to slightly increase the propagation length of the mode by stimulated emission of plasmon. We analyse the amplification phenomenon in the visible range by combining leakage radiation microscopy and surface plasmon coupled emission techniques.

Visual characterisation of TCNFs has been commonly performed using Scanning Electron Microscopy (SEM). Two-photon excitation microscopy of tryptophan-containing proteins.

We have examined the feasibility of observing single protein molecules by means of their intrinsic tryptophan emission after two-photon excitation. A respiratory protein from spiders, the 24-meric hemocyanin, containing 148 tryptophans, was studied in its native state under almost in vivo conditions. In this specific case, the intensity of the tryptophan emission signals the oxygen load, allowing one to investigate molecular cooperativity. As a system with even higher tryptophan content, we also investigated latex spheres covered with the protein avidin, resulting in 340 tryptophans per sphere.
