KIR2DL3 and the KIR ligand groups HLA-A-Bw4 and HLA-C2 predict the outcome of hepatitis B virus infection.

2017

Killer immunoglobulin-like receptors (KIRs) regulate the activation of Natural Killer cells through their interaction with human leukocyte antigens (HLA). KIR and HLA loci are highly polymorphic and certain HLA-KIR combinations have been found to protect against viral infections. In this study we analyzed whether the KIR/HLA repertoire may influence the course of hepatitis B virus (HBV) infection. Fifty-seven subjects with chronic hepatitis B (CHB), 44 subjects with resolved HBV infection, and 60 healthy uninfected controls (HC) were genotyped for KIR and their HLA ligands. The frequency of the HLA-A-Bw4 ligand group was higher in CHB (58%) than subjects with resolved infection (23%) (crude…

KRAS mutations testing in non-small cell lung cancer: the role of Liquid biopsy in the basal setting

2020

In advanced stage non-small cell lung cancer (NSCLC) patients, Kirsten Rat Sarcoma Viral Oncogene Homolog (KRAS) testing may soon acquire a predictive significance to select patients for AMG510 treatment. Since tissue samples are not always available, liquid biopsy may represent a viable option for KRAS testing. Here, we review the last three years clinical practice performed on 194 plasma based liquid biopsies by next generation sequencing (NGS) SiRe(®) panel. In particular, 36 (18.6%) KRAS mutated cases were identified, with an overall median allelic frequency of 5.0% (ranging between 0.2% and 46.8%). No concomitant mutations were observed in the other NSCLC clinical relevant genes includ…

Combination of the novel farnesyltransferase inhibitor RPR130401 and the geranylgeranyltransferase-1 inhibitor GGTI-298 disrupts MAP kinase activatio…

1999

To test the Kirsten-Ras (Ki-Ras) alternative prenylation hypothesis in malignant transformation, we used a novel farnesyltransferase inhibitor competitive to farnesyl-pyrophosphate, RPR130401, and a CaaX peptidomimetic geranylgeranyltransferase-1 inhibitor GGTI-298. In Ki-Ras-overexpressing transformed adrenocortical cells, RPR130401 at 1-10 microM inhibited very efficiently the [(3)H]farnesyl but not [(3)H]geranylgeranyl transfer to Ras. However, proliferation of these cells was only slightly sensitive to RPR130401 (IC(50)=30 microM). GGTI-298 inhibited the growth of these cells with an IC(50) of 11 microM but cell lysis was observed at 15 microM. The combination of 10 microM RPR130401 and…

HLA and killer cell immunoglobulin-like receptor (KIRs) genotyping in patients with acute ischemic stroke

2019

Abstract Introduction In humans, a major component of natural killer (NK) and T cell target recognition depends on the surveillance of human leukocyte antigen (HLA) class I molecules by killer immunoglobulin-like receptors (KIRs). Aims To implement the knowledge about the immunological genetic background of acute ischemic stroke susceptibility in relation to the frequency of the KIR genes and HLA alleles. Methods Subjects with acute ischemic stroke and subjects without stroke were genotyped for the presence of KIR genes and of the three major KIR ligand groups, HLA-C1, HLA-C2, and HLA-Bw4, both HLA-B and HLA-A loci. Results Between November 2013 and February 2016, consecutive patients with …

Kirsten ras mutations in patients with colorectal cancer: the 'RASCAL II' study

2001

Researchers worldwide with information about the Kirsten ras (Ki-ras) tumour genotype and outcome of patients with colorectal cancer were invited to provide that data in a schematized format for inclusion in a collaborative database called RASCAL (The Kirsten ras in-colorectal-cancer collaborative group). Our results from 2721 such patients have been presented previously and for the first time in any common cancer, showed conclusively that different gene mutations have different impacts on outcome, even when the mutations occur at the same site on the genome. To explore the effect of Ki-ras mutations at different stages of colorectal cancer, more patients were recruited to the database, whi…

Lajianalyysi ja valmennuksen ohjelmointi maastopyöräilyn olympialajissa (xco)

2013

Johdanto. Huippu-urheilijalta vaaditaan fyysisten ominaisuuksien lisäksi vahvaa henkistä kapasiteettia kestää vuosia jatkuvaa kovaa harjoittelua. Kilpapyöräily on yksi maailman seuratuimmista ja kilpailluimmista lajeista, joten huipputasolla fyysiset suorituskykyvaatimukset ovat korkeita Fysiologia. Maastopyöräilykilpailussa suurin osa kilpailuajasta ajetaan lähellä anaerobista kynnystä. Kovalla intensiteetillä ajettavassa kilpailussa miesammattilaiset tuottavat keskimäärin 360W tehon. Raskaissa maastoissa ajettavat kilpailut vaativat ajalta hyvää hapenottokykyä, siksi ammattilaismaastopyöräilijöiltä onkin mitattu varsin kovia jopa 79ml/kg/min vastaavia lukemia. Pyöräilyssä kehonpainolla on…

Kinematics of the ulna during pronation and supination in a cadaver study: implications for elbow arthroplasty

2003

Objective. Aim of this study was to exactly describe and quantify kinematics of the ulna during pro- and supination. Design. Biomechanical study in fresh frozen cadavers. Background. A previous MRI study revealed a varus/valgus motion of the ulna averaging 7.1° during pro-/supination. Axial rotation, however, could not be quantified. Methods. Sixteen arms were examined in a new apparatus that fixed the humerus on a template and allowed forearm rotation. Motion of a Kirschner wire placed in the ulna was recorded in steps of 30° by two perpendicularly arranged charge coupled device cameras during pro- and supination. Results. From supination to pronation the ulna showed a semi-lunar evasive m…
