Growth factors and cell kinetics: a mathematical model applied to Il-3 deprivation on leukemic cell lines.

1992

We assume the existence of a specific G1 protein which is an initiator of DNA replication. This initiator is supposed to be synthesized according to Michaelis-Menten kinetics. In order to start DNA replication, it is assumed that this G1 specific protein must be produced in a required amount. Intra-cellular growth inhibitors and extra-cellular growth factors control the production of the initiator. This model allows to calculate the average G1 phase time as a function of the various chemical concentrations of nutrients, enzymes, growth inhibitors and growth factors. This model is compared to cell kinetics experiments on a leukemic cell line responding to Interleukin 3 deprivation. The curve…

Review of Pharmacokinetics and Pharmacogenetics in Atypical Long-Acting Injectable Antipsychotics

2021

Over the last two decades, pharmacogenetics and pharmacokinetics have been increasingly used in clinical practice in Psychiatry due to the high variability regarding response and side effects of antipsychotic drugs. Specifically, long-acting injectable (LAI) antipsychotics have different pharmacokinetic profile than oral formulations due to their sustained release characteristics. In addition, most of these drugs are metabolized by CYP2D6, whose interindividual genetic variability results in different metabolizer status and, consequently, into different plasma concentrations of the drugs. In this context, there is consistent evidence which supports the use of therapeutic drug monitoring (TD…

Production and characterization of the recombinant Sphingomonas chlorophenolica pentachlorophenol 4-monooxygenase.

2001

Abstract Pentachlorophenol 4-monooxygenase (PCP4MO) from Sphingomonas chlorophenolica is a flavoprotein that hydroxylates PCP in the presence of NADPH and oxygen. In order to investigate the structure and function of active site, recombinant PCP4MO (rePCP4MO) was produced in Escherichia coli as a glutathione S-transferase (GST) fusion protein. Moreover, a tobacco etch virus (TEV) protease cleavage site (EKLYFQG) was introduced into GST-PCP4MO and a his-tagged TEV protease was employed. Hence, a two-step purification protocol was developed which allowed obtaining 15–20 mg of rePCP4MO from 1 L culture. The rePCP4MO revealed identity with native enzyme by SDS–PAGE and N-terminal sequence analy…

Kinetic properties of catecholoxidase activity of tarantula hemocyanin

2008

Phenoloxidases occur in almost all organisms, being essentially involved in various processes such as the immune response, wound healing, pigmentation and sclerotization in arthropods. Many hemocyanins are also capable of phenoloxidase activity after activation. Notably, in chelicerates, a phenoloxidase has not been identified in the hemolymph, and thus hemocyanin is assumed to be the physiological phenoloxidase in these animals. Although phenoloxidase activity has been shown for hemocyanin from several chelicerate species, a characterization of the enzymatic properties is still lacking. In this article, the enzymatic properties of activated hemocyanin from the tarantula Eurypelma californi…

Pharmacokinetics of new oral anticoagulants: implications for use in routine care

2018

Introduction: Since 2008, new oral anticoagulants (NOACs) have been approved for the prevention of venous thromboembolism (VTE) in patients receiving hip or knee replacement surgery, prevention of stroke and systemic embolism in patients with non-valvular atrial fibrillation (NVAF), treatment of deep vein thrombosis (DVT), and pulmonary embolism (PE). Premarketing randomized clinical trials (RCTs) of NOACs demonstrated their non-inferiority in terms of efficacy vs. warfarin (traditional oral anticoagulant–TOA), with lower risk of serious adverse drug reactions, especially cerebral hemorrhages. In clinical practice, pharmacokinetic aspects of NOACs have to be carefully taken into account to …

Ocena właściwości sorpcyjnych Hottonia palustris L.

2016

Zbadano w warunkach laboratoryjnych kinetykę i równowagę procesu sorpcji wybranych metali ciężkich: Cu i Zn w roślinie wodnej - okrężnicy bagiennej (Hottonia palustris L.). Do opisu równowag wykorzystano model izotermy Langmuira. Stwierdzono, że Hottonia palustris L. sorbuje jony metali ciężkich proporcjonalnie do ich zawartości w roztworze, w którym została zanurzona. Na podstawie przeprowadzonych badań można przypuszczać…

Particle models for kinetic equations: an introduction and some rigorous results

2011

We shall give an introduction to the validity problem for kinetic equations and we shall review some convergence theorems concerning the derivation from a microscopic dynamics of systems of partial differential equations describing, at the mesoscopic scale, collections of particles interacting through various (collisions and mean field) type of interaction.

Revisiting the disappearance of terrestrial dissolved organic matter in the ocean: A δ13C study

2014

Abstract. Organic carbon (OC) depleted in 13 C is a widely used tracer for terrestrial organic matter (OM) in aquatic sys- tems. Photochemical reactions can, however, change δ 13 C of dissolved organic carbon (DOC) when chromophoric, aromatic-rich terrestrial OC is selectively mineralized. We assessed the robustness of the δ 13 C signature of DOC ( δ 13 C DOC ) as a tracer for terrestrial OM by estimating its change during the photobleaching of chromophoric DOM (CDOM) from 10 large rivers. These rivers cumulatively ac- count for approximately one-third of the world’s freshwater discharge to the global ocean. Photobleaching of CDOM by simulated solar radiation was associated with the photoch…

Application of computational fluid dynamics technique in electrodialysis/reverse electrodialysis processes

2022

This Chapter treats the application of CFD technique in ED/RED processes. CFD simulations are able to predict flow and mass transfer in channels filled with spacers or membrane profiles, by providing dimensionless correlations useful for process models. At a larger scale, manifolds of the stack and inlet/ outlet zones of the channel can be simulated. Numerical models can include the computation of the electrical potential, thus characterizing the process performance. Direct numerical simulations can elucidate the complex phenomenology of overlimiting transport and of chaotic electrokinetic flows.

Significant interaction between high-dose methotrexate and high-dose piperacillin-tazobactam causing reversible neurotoxicity and renal failure in an…

2020

Introduction Pharmacokinetic interaction of high-dose methotrexate (MTX) and other concomitantly administered renally secreted medicinal products may lead to insufficient methotrexate serum level decrease and significant MTX toxicity. Case report We report the case of an 18-year-old male patient treated with high-dose MTX for an osteosarcoma and with high-dose piperacillin-tazobactam at the same time. MTX serum levels were severely elevated 24 hours after the MTX infusion and did not decrease in accordance with the specific calcium folinate rescue protocol. The patient experienced renal failure accompanied by neurological symptoms, most consistent with MTX-related renal and CNS toxicity. Ma…
