A multicenter phase 1 study of solitomab (MT110, AMG 110), a bispecific EpCAM/CD3 T-cell engager (BiTE®) antibody construct, in patients with refract…
2018
ABSTRACT We assessed the tolerability and antitumor activity of solitomab, a bispecific T-cell engager (BiTE®) antibody construct targeting epithelial cell adhesion molecule (EpCAM). Patients with relapsed/refractory solid tumors not amenable to standard therapy received solitomab as continuous IV infusion in a phase 1 dose-escalation study with six different dosing schedules. The primary endpoint was frequency and severity of adverse events (AEs). Secondary endpoints included pharmacokinetics, pharmacodynamics, immunogenicity, and antitumor activity. Sixty-five patients received solitomab at doses between 1 and 96 µg/day for ≥28 days. Fifteen patients had dose-limiting toxicities (DLTs): e…
Bone targeting compounds for radiotherapy and imaging: *Me(III)-DOTA conjugates of bisphosphonic acid, pamidronic acid and zoledronic acid
2016
Background Bisphosphonates have a high adsorption on calcified tissues and are commonly used in the treatment of bone disorder diseases. Conjugates of bisphosphonates with macrocyclic chelators open new possibilities in bone targeted radionuclide imaging and therapy. Subsequent to positron emission tomography (PET) examinations utilizing 68Ga-labelled analogues, endoradiotheraphy with 177Lu-labelled macrocyclic bisphosphonates may have a great potential in the treatment of painful skeletal metastases. Methods Based on the established pharmaceuticals pamidronate and zoledronate two new DOTA-α-OH-bisphosphonates, DOTAPAM and DOTAZOL(MM1.MZ) were successfully synthesized. The ligands were labe…
Evaluation of the inverse electron demand Diels-Alder reaction in rats using a scandium-44-labelled tetrazine for pretargeted PET imaging
2019
Background Pretargeted imaging allows the use of short-lived radionuclides when imaging the accumulation of slow clearing targeting agents such as antibodies. The biotin-(strept)avidin and the bispecific antibody-hapten interactions have been applied in clinical pretargeting studies; unfortunately, these systems led to immunogenic responses in patients. The inverse electron demand Diels-Alder (IEDDA) reaction between a radiolabelled tetrazine (Tz) and a trans-cyclooctene (TCO)-functionalized targeting vector is a promising alternative for clinical pretargeted imaging due to its fast reaction kinetics. This strategy was first applied in nuclear medicine using an 111In-labelled Tz to image TC…
Kinetics of photochemical isomerization of TFA-Gly-ZΔPhe into TFA-Gly-EΔPhe
2017
The kinetics of photoisomerization of trifluoroacetyl-(Z)-dehydrophenylalanylglycine into trifluoroacetyl-(E)- dehydrophenylalanylglycine was studied in the hope that light-induced reaction could be useful as a means of preparation of the E-dehydropeptides. The obtained results indicate that if this reaction carried out under irradiation with light of wavelength 360 nm it is practically irreversible and gave nearly quantitatively pure Eisomer Significantly, expected cyclic side-products were not observed in the reaction mixture, thus proving the preparative potential of the elaborated procedure.
Distribution and depletion of sulfadiazine after a multiple per os dosing in gilthead sea bream (Sparus aurata) fed two different diets
2013
The distribution and depletion profile of sulfadiazine (SDZ) were investigated in gilthead sea bream (Sparus aurata) fed on fish oil (FO) or plant oil-based (PO) diets. Fish averaging 230 g were given medicated feed containing 25 mg SDZ kg-1 fish for 5 days at 24-26°C. Blood and muscle plus skin were sampled on days 1, 3, 5, 6, 8 and 9. Differences in plasma and fillet SDZ levels between the two groups were statistically insignificant. The maximum drug concentrations in plasma were 3.2 ± 1.9 μg mL-1 and 2.9 ± 1.2 μg mL-1 in the PO and FO groups, respectively. In post-medicated samples depletion rapidly reached concentrations close to the level of quantification at 72 h post medication. With…
Validation of 2D Force Measurement Roller Ski and Practical Application.
2022
Several methods could be used to measure the forces from skis or roller skis in cross-country skiing. Equipment that could measure medio-lateral forces may be of good help for investigating the relevant skating techniques. The aim of this study was to validate a pair of newly designed two-dimensional force measurement roller skis. The vertical and medio-lateral forces which were perpendicular to the body of the roller ski could be measured. Forces were resolved into the global coordinate system and compared with the force components measured by a force plate. A static and dynamic loading situation for the force measurement roller ski was performed to reveal the validity of the system. To de…
Detailed modeling of the kraft pulping chemistry : carbohydrate reactions
2020
The article introduces a detailed model for carbohydrate chemistry in kraft pulping. This article is continuation to the modeling work carried out for hot water extraction and chemical pulp bleaching. The model includes galactoglucomannan, xylan, and cellulose acid–base equilibria, in addition to peeling, stopping, and alkaline hydrolysis reactions of the same carbohydrates, as well as hexenuronic acid formation and degradation reactions. The Arrhenius parameters were applied from the literature or regressed against experimental data in the present study. The model is very successful in predicting the experimental data of carbohydrate reactions during kraft pulping. Many features of the pul…
Modeling biomass char gasification kinetics for improving prediction of carbon conversion in a fluidized bed gasifier
2014
Gasification of biomass in a fluidized bed (FB) was modeled based on kinetic data obtained from previously conducted thermo- gravimetic analysis. The thermogravimetric analysis experiments were designed to closely resemble conditions in a real FB gasifier by using high sample heating rates, in situ devolatilization and gas atmospheres of H 2 O / H 2 and CO 2 / CO mixtures. Several char kinetic models were evaluated based on their ability to predict char conversion based on the thermogravimetric data. A modified version of the random pore model was shown to provide good fitting of the char reactivity and suitability for use in a reactor model. An updated FB reactor model which incorporates t…
Optical Studies of the Capillary Refill Kinetics in Fingertips
2009
A new methodology for temporal analysis of the capillary refill processes by means of photoplethysmography (PPG) contact probe operating in the blue region of spectrum has been developed. A corresponding prototype device for finger measurements has been created and clinically tested. Results demonstrated that both AC and DC components of the blue PPG bio-signals are sensitive to the capillary occlusion and refill. Real-time measurements of the skin capillary refill kinetics by this technology can be used for noninvasive diagnostics of the peripheral perfusion disorders.
Essential Oil Composition of Alluaudia procera and in Vitro Biological Activity on Two Drug-Resistant Models
2019
Drug resistance is a major obstacle in antibiotic and antitumor chemotherapy. In response to the necessity to find new therapeutic strategies, plant secondary metabolites including essential oils (EOs) may represent one of the best sources. EOs in plants act as constitutive defenses against biotic and abiotic stress, and they play an important role in the pharmacology for their low toxicity, good pharmacokinetic and multitarget activity. In this context, natural products such as EOs are one of the most important sources of drugs used in pharmaceutical therapeutics. The aim of this paper was to identify the chemical composition of the essential oil of Alluaudia procera leaves, obtained by hy…
