The main objective of this study was to determine the prevalence of the Qnr determinants in clinical and environmental Aeromonas spp. A total of 52 Aeromonas sp. isolates identified by biochemical methods (5), 25 isolated from natural waters (1) and 27 isolated from clinical samples from hospitals in Valencia, Spain, were tested for quinolone resistance by the disk diffusion method (4) (nalidixic acid, 30 μg; oxolinic acid, 2 μg; flumequine, 30 μg; ciprofloxacin, 5 μg; and levofloxacin, 5 μg). By the enrichment culture technique 14 gram-negative bacteria and two yeast strains were isolated that used D(+)-malic acid as sole carbon source. The bacteria were identified as Pseudomonas putida, Pseudomonas fluorescens, Pseudomonas aeruginosa and Klebsiella aerogenes. In cell-free extracts of P. fluorescens and P. putida the presence of malate dehydrogenase, D-malic enzyme (NAD-dependent) and L-malic enzyme (NADP-dependent) was demonstrated. Summary The faecal coliforms at different sites of a hypereutrophic lake near Valencia (Albufera) were identified and their relative amounts established along an annual cycle. Using lauryl tryptose broth at 35°C, followed by incubation at 44.4°C in 2% brilliant green bile, Escherichia coli and Klebsiella pneumoniae are practically the only coliforms present. A positive correlation was found between the water temperature and the relative amount of these two coliforms: K. pneumoniae predominates at high water temperatures, whereas E. coli shows preponderance during the cold period.
