Expression and purification of polyhistidine-tagged firefly luciferase in insect cells

The coleopteran firefly, Photinus pyralis, luciferase was produced in lepidopteran Trichoplusia ni insect cells using a baculovirus expression vector. The recombinant protein was equipped with a polyhistidine affinity tag at the carboxyl terminus and purified by immobilized metal-ion affinity chromatography in combination with an expanded bed adsorption system. This approach enabled an efficient, one-step purification protocol of a genetically modified luciferase with properties similar to those of the authentic counterpart. Pachydema lopadusanorum n.sp. (Coleoptera Melolonthidae) from Lampedusa Island (Sicily Channel, Central Metditerranean Sea, Italy)

In this paper the populations of Pachydema Castelnau, 1832 (Coleoptera Melolonthidae) living in Lampedusa Island (Sicily Channel, Central Mediterranean Sea, Italy), so far attributed to P. hirticollis (Fabricius, 1787) of North Africa, are examined. The comparison of the main morphological characters between these two populations allowed to attribute those of Lampedusa to a new species that is described in the present work. Faunistic and biological observations on these species are provided.
The role of lantern fish (Myctophidae) in the life-cycle of cetacean parasites from western Mediterranean waters

Myctophids (lantern fish) and cephalopods play a key role in trophic webs from the continental slope and oceanic waters linking the zooplankton to top predators. Many cetaceans feed on both lantern fish and cephalopods, and such prey would thus be expected to bridge the trophic gap in the life-cycles of helminths infecting cetaceans. However, information on the life-cycles of most of these helminths is extremely scanty. The volume of boat traffic and its potential connection to the coastal distribution of the common bottlenose dolphin (Tursiops truncatus) was evaluated off Lampedusa Island (Strait of Sicily). From July to September 2006 daily surveys were carried out at eight sites along the coast, three times a day, to assess the number, type, and size of boats moving, fishing, or stationed in Lampedusa waters. The study area was divided into four geographic areas: northwest, northeast, southwest, and southeast. Sequence-specific and DNA structure-dependent interactions of Escherichia coli MutS and human p53 with DNA

Many proteins involved in DNA repair systems interact with DNA that has structure altered from the typical B-form helix. Using magnetic beads to immobilize DNAs containing various types of structures, we evaluated the in vitro binding activities of two well-characterized DNA repair proteins, Escherichia coli MutS and human p53. E. coli MutS bound to double-stranded DNAs, with higher affinity for a G/T mismatch compared to a G/A mismatch and highest affinity for larger non-B-DNA structures. E. coli MutS bound best to DNA between pH 6 and 9. Iodine and mercury resonance lamps for kinetics experiments and their spectra in the far ultraviolet

Electrodeless iodine and mercury iodide radio-frequency discharge lamps have been made to provide intense sources of resonance radiation of iodine and mercury in the 183-253 nm region. Production technology and emission spectra in the ultraviolet and vacuum ultraviolet regions are described. Both lamps (iodine and mercury iodide) are designed and used for monitoring the iodine 2P1/2 (206.163 nm) and 2P3/2 (183.038 nm) level population kinetics after flash photolysis of I2 or other precursor molecules in laboratory experiments. The technique is suitable for the investigation of the gas phase reaction kinetics following the photolysis of I2 in the presence of O3.

1,4-dihydropyridines (1,4-DHP) possess important biochemical and pharmacological properties, including antioxidant and antimutagenic activities. Interaction of some 1,4-DHP with DNA was recently reported. AV-153-Na, an antimutagenic and DNA-repair-enhancing compound appeared to be able to interact with DNA by intercalation.
