GABAA-receptor Subtypes: Clinical Efficacy and Selectivity of Benzodiazepine Site Ligands

1997

The main inhibitory neurotransmitter receptor of the brain, the gamma-aminobutyric acid type A receptor (GABA[A]), mediates the actions of several classes of clinically important drugs, such as benzodiazepines, barbiturates and general anaesthetics. This review summarizes the current knowledge on how classical benzodiazepines and novel nonbenzodiazepine compounds act on the benzodiazepine site of GABA(A) receptors and on their clinical pharmacology related to anxiolytic, sedative, hypnotic and cognitive effects or side-effects. Partial agonism, receptor subtype selectivity and novel binding sites are discussed as possible strategies to develop new drugs with fewer adverse effects than are s…

Zaleplon displays a selectivity to recombinant GABAA receptors different from zolipdem, zopiclone and benzodiazepines

1999

A chemically heterogeneous group of compounds acts at the benzodiazepine (BZ) recognition site of the γ-aminobutyric acid type A (GABAA) receptor complex. Whereas most 1,4-BZs recognize all GABAA/BZ receptors with similar affinity, other compounds differentiate between the large number of native GABAA receptors which assemble from the more than 14 known subunits. Here we describe the in vitro binding properties of the BZs lorazepam and Ro 15-4513 plus the three hypnotics zaleplon, zolpidem and zopiclone to eight receptors subtypes. Lorazepam fits well into the general shceme for other 1,4-BZs with respect to its receptor subtype selectivity in spite of its clinically different use. Zaleplon…

Evaluation of cropping systems for management of herbicide-resistant populations of blackgrass (Alopecurus myosuroides Huds.)

2001

Abstract Simplification of cropping systems often leads to an increase in weed populations which require an intensive use of herbicides to maintain populations at an acceptable level. Due to a heavy reliance on herbicides and a lack of cultural control measures, herbicide-resistant blackgrass (Alopecurus myosuroides Huds.) biotypes appeared recently in France. An experiment was conducted to evaluate the effects of different cropping systems on a population of herbicide-resistant blackgrass. Two crop rotations, one consisting exclusively of winter crops and another including spring crops, were assessed over a three-year period. Crop rotation was combined with different cultural practices (mo…

Assessing non-chemical weeding strategies through mechanistic modelling of blackgrass (Alopecurus myosuroides Huds.) dynamics

2010

; Because of environmental and health safety issues, it is necessary to develop strategies that do not rely on herbicides to manage weeds. Introducing temporary grassland into annual crop rotations and mechanical weeding are the two main features that are frequently used in integrated and organic cropping systems for this purpose. To evaluate the contribution of these two factors in interaction with other cropping system components and environmental conditions, the present study updated an existing biophysical model (i.e. AlomySys) that quantifies the effects of cropping system on weed dynamics. Based on previous experiments, new sub-models were built to describe the effects on plant survi…
