Functional Role of Lipoprotein Receptors in Alzheimers Disease

2008

The LDL receptor gene family constitutes a class of structurally closely related cell surface receptors fulfilling diverse functions in different organs, tissues, and cell types. The LDL receptor is the prototype of this family, which also includes the VLDLR, ApoER2/LRP8, LRP1 and LRP1B, as well as Megalin/GP330, SorLA/LR11, LRP5, LRP6 and MEGF7. Recently several lines of evidence have positioned the LDL receptor gene family as one of the key players in Alzheimer's disease (AD) research. Initially this receptor family was of high interest due to its key function in cholesterol/apolipoprotein E (ApoE) uptake, with the epsilon4 allele of ApoE as the strongest genetic risk factor for late-onse…

Functional role of the low-density lipoprotein receptor-related protein in Alzheimer's disease.

2006

Alzheimer’s disease (AD) is the most common age-related neurodegenerative disorder, characterized by neuronal loss, neurofibrillary tangle formation and the extracellular deposition of amyloid-β (Aβ) plaques. The amyloid precursor protein (APP) and the enzymes responsible for Aβ generation seem to be the base elements triggering the destructive processes. Initially, the low-density lipoprotein receptor-related protein (LRP) was genetically linked to AD and later it emerged to impact on many fundamental events related to this disease. LRP is not only involved in Aβ clearance but is also the major receptor of several AD-associated ligands, e.g. apolipoprotein E and α2-m…

Alzheimer’s disease and genetics of inflammation: a pharmacogenomic vision

2007

Inflammation plays a key role in Alzheimer disease, and dissecting the genetics of inflammation may provide an answer to the possible treatment. The next-generation therapy is based on a pharmacogenomics that will reconure new approaches to a drug used on definite people with specific dosage. The translation of pharmacogenomics into clinical practice will allow bold steps to be taken toward personalized medicine. In response to tissue injury elicited by trauma or infection, the inflammatory response sets in as a complex network of molecular and cellular interactions, directed to facilitate a return to physiological homeostasis and tissue repair. The role of an individual’s genetic backgroun…

Variability of the African convection centre as viewed by outgoing longwave radiation records and relationships with sea-surface temperature patterns

1995

International audience; The main African convection centre (ACC) is delimited by the 5"-square areas with outgoing longwave radiation less than 235 W m-' in the domain 17.5°N-17.5"S and 18"W-46"E. Four parameters (latitude and longitude of the centre of gravity, mean intensity, and extension) were analysed during the period June 1974 to December 1991. The relationships between the variability of the ACC and the main sea-surface temperature (SST) patterns were then studied. Correlation and composite analyses demonstrate that the first principal component of SST, representing mainly the variability of the eastern and the central tropical Pacific, is associated strongly with the extension and …

Effect of temperature and traffic on mix-design of bituminous asphalt for railway sub-ballast layer

2017

A wide integrated research has been carried out to assess the suitability of bituminous mixtures as the sub-ballast layer in railways. The influence of temperature on the mechanical characteristics and thermal susceptibility of the bituminous conglomerate—which forms sub-ballast layer of railway lines—has provided the motivation for carrying out a measurement of the thermal cycles in this layer. The evaluation of the average seasonal temperatures has proven to be well interpolated by sinusoidal functions, of which characteristic parameters are determined. To expand the validity of the study in different weather situations apart from Sicily, where the experiment was held, we proceeded to app…

Petrology and geochemistry of submarine volcanism in the Sicily Channel Rift

2006

Submarine magmatism in the Sicily Channel Rift began in the early Pliocene and lasted until almost 200 yr ago. We present here petrological and geochemical data on volcanic rocks dredged from Graham and Nameless banks and Pantelleria seamounts in the Pelagian sector of the Sicily Channel Rift. Petrological evidence suggests that the ascent of magmas to the surface was relatively rapid, probably through channels superimposed over the major tectonic discontinuities of the Rift. Major and trace element data indicate an ocean island basalt affinity for Graham and Nameless bank alkaline lavas and a depleted tholeiitic signature for one Pantelleria seamount, which had a shallower mantle source. S…

Wnt activity defines colon cancer stem cells and is regulated by the microenvironment.

2010

Despite the presence of mutations in APC or beta-catenin, which are believed to activate the Wnt signalling cascade constitutively, most colorectal cancers show cellular heterogeneity when beta-catenin localization is analysed, indicating a more complex regulation of Wnt signalling. We explored this heterogeneity with a Wnt reporter construct and observed that high Wnt activity functionally designates the colon cancer stem cell (CSC) population. In adenocarcinomas, high activity of the Wnt pathway is observed preferentially in tumour cells located close to stromal myofibroblasts, indicating that Wnt activity and cancer stemness may be regulated by extrinsic cues. In agreement with this noti…
