Purkinje cell loss and motor coordination defects in profilin1 mutant mice.

2012

Profilin1 is an actin monomer-binding protein, essential for cytoskeletal dynamics. Based on its broad expression in the brain and the localization at excitatory synapses (hippocampal CA3-CA1 synapse, cerebellar parallel fiber (PF)-Purkinje cell (PC) synapse), an important role for profilin1 in brain development and synapse physiology has been postulated. We recently showed normal physiology of hippocampal CA3-CA1 synapses in the absence of profilin1, but impaired glial cell binding and radial migration of cerebellar granule neurons (CGNs). Consequently, brain-specific inactivation of profilin1 by exploiting conditional mutants and Nestin-mediated cre expression resulted in a cerebellar hyp…

Dual effects of increased glycogen synthase kinase-3β activity on adult neurogenesis

2013

Adult neurogenesis, the generation of new neurons during the adulthood, is a process controlled by several kinases and phosphatases among which GSK3β exerts important functions. This protein is particularly abundant in the central nervous system, and its activity deregulation is believed to play a key role in chronic disorders such as Alzheimer's disease. Previously, we reported that in vivo overexpression of GSK3β (Tet/GSK3β mice) causes alterations in adult neurogenesis, leading to a depletion of the neurogenic niches. Here, we have further characterized those alterations, finding a delay in the switching-off of doublecortin marker as well as changes in the survival and death rates of imm…

Permanent plastid — nuclear complexes (PNCs) in plant cells

2008

Conventional opinion assumes random distribution of plastids in the plant cell and light regulated movement realised with a help of stromules and actin microfilaments. In several organisms from protists to plants the joining of chloroplasts to the nucleus has been mentioned as a phenomenon [1–3]. However, little is known what plants and tissues and how frequently contain these structures? Whether appearance of PNC in cell depends on the state of differentiation? What is physiologic role of the PNC.

Characterization of the interaction between Actinin-Associated LIM Protein (ALP) and the rod domain of α-actinin

2009

Abstract Background The PDZ-LIM proteins are a family of signalling adaptors that interact with the actin cross-linking protein, α-actinin, via their PDZ domains or via internal regions between the PDZ and LIM domains. Three of the PDZ-LIM proteins have a conserved 26-residue ZM motif in the internal region, but the structure of the internal region is unknown. Results In this study, using circular dichroism and nuclear magnetic resonance (NMR), we showed that the ALP internal region (residues 107–273) was largely unfolded in solution, but was able to interact with the α-actinin rod domain in vitro, and to co-localize with α-actinin on stress fibres in vivo. NMR analysis revealed that the ti…

Aerobic granular sludge treating high strength citrus wastewater: Analysis of pH and organic loading rate effect on kinetics, performance and stabili…

2017

Abstract In the present paper, the feasibility of citrus wastewater treatment with aerobic granular sludge sequencing batch reactors (AGSBR) was investigated. Two AGSBRs (named R1 and R2, respectively) were operated for 90 days under different organic loading rates (OLR) and pH in two experimental periods. The OLR ranged approximately between 3.0 kg TCOD m−3d−1 and 7 kg TCOD m−3d−1 during Period I, whereas between 7 kg TCOD m−3d−1 and 15 kg TCOD m−3d−1 during Period II. pH was maintained at 7.0 and 5.5 in R1 and R2, respectively. The results revealed that under high OLR and unbalanced feast/famine regime (Period I), the development of fast-growing microorganisms (fungi and filamentous bacte…
