Gas cell studies of thorium using filament dispensers at IGISOL

Abstract Filament-based dispensers of thorium have been investigated at the IGISOL facility, Jyvaskyla, for potential use as a thorium ion source for future collinear laser spectroscopy experiments. Several different filaments were manufactured in the Institute of Atomic and Subatomic Physics of TU Wien, with 232Th and 229Th prepared on tantalum substrates either by drying thorium nitrate solution or via molecular plating, while adding a layer of zirconium for oxide reduction. Myosin VIIa, harmonin and cadherin 23, three Usher I gene products that cooperate to shape the sensory hair cell bundle

Deaf-blindness in three distinct genetic forms of Usher type I syndrome (USH1) is caused by defects in myosin VIIa, harmonin and cadherin 23. Despite being critical for hearing, the functions of these proteins in the inner ear remain elusive. Here we show that harmonin, a PDZ domain-containing protein, and cadherin 23 are both present in the growing stereocilia and that they bind to each other. Moreover, we demonstrate that harmonin b is an F-actin-bundling protein, which is thus likely to anchor cadherin 23 to the stereocilia microfilaments, thereby identifying a novel anchorage mode of the cadherins to the actin cytoskeleton. Detection of cytokeratin dynamics by time-lapse fluorescence microscopy in living cells.

To monitor the desmosome-anchored cytokeratin network in living cells fusion protein HK13-EGFP consisting of human cytokeratin 13 and the enhanced green fluorescent protein was stably expressed in vulvar carcinoma-derived A-431 cells. It is shown for A-431 subclone AK13-1 that HK13-EGFP emits strong fluorescence in fixed and living cells, being part of an extended cytoplasmic intermediate filament network that is indistinguishable from that of parent A-431 cells.
