Thermal Denaturation of Pea Globulins (Pisum sativum L.)—Molecular Interactions Leading to Heat-Induced Protein Aggregation

The heat-induced denaturation and aggregation of mixed pea globulins (8%, w/w) were investigated using differential scanning calorimetry (DSC), SDS-PAGE, and size-exclusion chromatography (SEC-HPLC). DSC data showed that the pea proteins denaturation temperature (T(d)) was heating-rate dependent. The T(d) value decreased by about 4 °C by lowering the heating rate from 10 to 5 °C/min. The SDS-PAGE analysis revealed that protein denaturation upon heating at 90 °C was mainly governed by noncovalent interaction. The SEC-HPLC measurements indicated that low-denatured legumin (≈350-410 kDa) and vicilin/convicilin (≈170 kDa) globulins were heat-denatured and most of their subunits reassociated int…

Nomenclatural notes on Fabaceae described from Sicily

A note about some taxa in Anthyllis and Trifolium (Fabaceae) described from Sicily is presented. Concerning Trifolium, we provide the lectotypification of the name T. pratense var. semipurpureum (≡ T. pratense subsp. semipurpureum), using a specimen preserved at PAL (Todaro’s collection). Concerning the Lojacono-Pojero’s name Vulneraria heterophylla var. parvula, it is lectotypified by a specimen at G. A neotype at NAP is proposed for the Gussone’s name Vulneraria heterophylla var. albiflora. As this latter name is applied to a taxon accepted here at specific rank, a new combination is validated. The biodiversity of nitrogen-fixing organisms is huge. Taxonomic and phylogenetic research is needed to structure this diversity, to facilitate communication among scientists, and to increase our understanding of the evolution and biology of diazotrophs. Molecular tools for taxonomic and biodiversity studies of diazotrophic rhizobia, frankiae, cyanobacteria and bacilli are presented in sections 2 to 5. Sections 6 to 9 focus on problems with genus and species assignment.

Rhizobium leguminosarum biovar viciae genotypes interact with pea plants in developmental responses of nodules roots and shoots

International audience; The variability of the developmental responses of two contrasting cultivars of pea (Pisum sativum) was studied in relation to the genetic diversity of their nitrogen-fixing symbiont Rhizobium leguminosarum bv. viciae. A sample of 42 strains of pea rhizobia was chosen to represent 17 genotypes predominating in indigenous rhizobial populations, the genotypes being defined by the combination of haplotypes characterized with rDNA intergenic spacer and nodD gene regions as markers.
