Quantitative analysis of the impact of a human pathogenic mutation on the CCT5 chaperonin subunit using a proxy archaeal ortholog

The human chaperonin complex is a ~ 1 MDa nanomachine composed of two octameric rings formed from eight similar but non-identical subunits called CCT. Here, we are elucidating the mechanism of a heritable CCT5 subunit mutation that causes profound neuropathy in humans. In previous work, we introduced an equivalent mutation in an archaeal chaperonin that assembles into two octameric rings like in humans but in which all subunits are identical. We reported that the hexadecamer formed by the mutant subunit is unstable with impaired chaperoning functions. Abstract In the present research, we study a possibility of using the electric spark erosion method as an alternative to the method of chemical co-precipitation for preparation of magnetic nanoparticles. Initiation of high frequency electric discharge between coarse iron particles under a layer of distilled water allows obtaining pure magnetite nanoparticles.

Immunomodulatory Therapy of Inflammatory Liver Disease Using Selectin-Binding Glycopolymers

Immunotherapies have the potential to significantly advance treatment of inflammatory disease and cancer, which are in large part driven by immune cells. Selectins control the first step in immune cell adhesion and extravasation, thereby guiding leukocyte trafficking to tissue lesions. We analyzed four different highly specific selectin-binding glycopolymers, based on linear poly(2-hydroxypropyl)-methacrylamide (PHPMA) polymers. These glycopolymers contain either the tetrasaccharide sialyl-LewisX (SLeX) or the individual carbohydrates fucose, galactose, and sialic acids mimicking the complex SLeX binding motive. Abstract If a light beam meets a fluid that contains scattering centers randomly distributed in suspension, light is scattered by each of them. If the light source is coherent, the scattered waves will be also coherent, therefore they will interfere. The fluctuations of the far-field interference signal, once recorded and digitized, become a time series that can be later on analyzed to produce the average size of the suspended particles or the size distribution. Glycerosomes: Use of hydrogenated soy phosphatidylcholine mixture and its effect on vesicle features and diclofenac skin penetration.

In this work, diclofenac was encapsulated, as sodium salt, in glycerosomes containing 10, 20 or 30% of glycerol in the water phase with the aim to ameliorate its topical efficacy. Taking into account previous findings, glycerosome formulation was modified, in terms of economic suitability, using a cheap and commercially available mixture of hydrogenated soy phosphatidylcholine (P90H). The synthesis and aggregation behaviour in aqueous media of novel amphiphilic poly(ethylene oxide)-block-poly(n-butyl acrylate)-block-poly(acrylic acid) (PEO–PnBA–PAA) triblock terpolymers were studied. Terpolymers composed of two highly asymmetric hydrophilic PEO (113 monomer units) and PAA (10–17 units) blocks, and a longer soft hydrophobic PnBA block (163 or 223 units) were synthesized by atom transfer radical polymerisation (ATRP) of n-butyl acrylate and tert-butyl acrylate (tBA), followed by selective hydrolysis of the PtBA blocks. Light scattering in artificial fog and simulated with light scattering filter.

Disability glare, affecting e.g. road safety at night, may result either from intraocular light scattering or from external conditions such as fog. Measurements were made of light scattering in fog and compared with intraocular straylight data for normal eyes and eyes with simulated cataract. All measurements were made with a direct compensation flicker method. To estimate light scattering levels in fog, straylight measurements were carried in a fog chamber for different densities of fog. Targeted Hybrid Nanocarriers as a System Enhancing the Skin Structure

The skin is constantly exposed to external and internal factors that disturb its function. In this work, two nanosystems-levan nanoparticles and a surfactin-stabilized nanoemulsion were preserved (tested for microbial growth) and characterized (size, polydispersity, Zeta potential, and stability). The nanosystems were introduced in the model formulations-cream, tonic, and gel, and confirmed by TEM. The analysis showed that nanoemulsion has a spherical morphology and size 220–300 nm, while levan nanoparticles had irregular shapes independently of the use of matrix and with particle size (130–260 nm).
