Expression and purification of polyhistidine-tagged firefly luciferase in insect cells

The coleopteran firefly, Photinus pyralis, luciferase was produced in lepidopteran Trichoplusia ni insect cells using a baculovirus expression vector. The recombinant protein was equipped with a polyhistidine affinity tag at the carboxyl terminus and purified by immobilized metal-ion affinity chromatography in combination with an expanded bed adsorption system. This approach enabled an efficient, one-step purification protocol of a genetically modified luciferase with properties similar to those of the authentic counterpart. In the last few years there has been an increasing attempt to find solutions on how to mitigate the environmental impacts of air travel. Behavioral change has hereby been identified as the measure with the most significant impact. One way to encourage behavioral change is the use of eco-labels. Eco-labels have, to date, received scant attention in the aviation industry, and their effect on air travel behavior is still largely unknown. Scent markings (urine and faeces) of small mammals are visible in ultraviolet (UV) light. Diurnal kestrels, Falco tinnunculususe them as a cue to find areas of food abundance. We studied whether vole-eating, nocturnal Tengmalm's owls, Aegolius funereuscan see vole scent marks using UV-vision. In a laboratory experiment, 14 young (less than 6 months old) and 14 adult (more than 6 months old) owls were individually given a choice between four adjacent arenas: (1) an arena with vole urine and faeces in UV light; (2) an arena with vole urine and faeces in visible light; (3) a clean arena in UV light; and (4) a clean arena in visible light. Summary Sinapis alba (white mustard) plants were grown under conditions of strong (100 W · m -2 ) and weak (3 W · m -2 ) light and the effect of the light intensity during growth on the CO 2 -compensation point and the activities of glycolic acid oxidase and ribulose bisphosphate carboxylase were studied. The density and movements of fish under ice were studied with single-beam mobile surveys and fixed location split-beam surveys, as well as exploratory fishing in winter-seining areas of two shallow Finnish lakes during winter 1999. Fish schooled near the bottom during the day but the schools dispersed and fish ascended at night. Single and split-beam target strength distributions corresponded fairly closely with the length distribution of seine catch samples. Estimated fish densities were greater at night than in daylight. Bird of Paradise tree (Strelitzia alba (L. f.) Skeels) is an ornamental perennial tropical plant grown in southern Italy. In the summer of 2006 and 2007, a widespread, severe leaf disease was observed on seedlings and 1- to 2-year-old plants in two glasshouses located in eastern Sicily. Disease incidence ranged from 10 to 25%. Symptoms on the leaves consisted of dark brown-to-black stripes of varying length and found between the lateral veins. Lesions sometimes coalesced into a large area of necrotic tissue. Symptomatic tissues were ground in a drop of sterile distilled water (SDW) with a scalpel. Medinilla magnifica Lindl., Mandevilla splendens (Hook.) Woodson, the hybrid Mandevilla × amoena ‘Alice du Pont’ (pink allamanda), and various species of Allamanda, such as A. cathartica L. and A. blanchetii A. DC. (purple allamanda), are grown in Sicily as ornamentals. After a frost in early December 2001, a sudden wilt of container-grown cuttings of these tropical species was observed in a plastic-covered production greenhouse, with ≈30% of M. magnifica plants and 70% of Mandevilla and Allamanda plants affected.
