Mapping and quantification of biomolecules in tumor biopsies using bioluminescence

Quantitative bioluminescence and single-photon imaging have been applied for mapping concentration distributions of metabolites, such as adenosine triphosphate (ATP), glucose and lactate, in biopsies of cervical cancers in patients. Biopsies were taken before a conventional radiation treatment, and a number of clinically relevant data, such as local tumor control, patient survival, metastatic spread and so forth, were documented. There was no correlation between staging or grading and any of the metabolic parameters measured. Comparative imaging of structure and metabolites in tumours.

A novel technique for metabolic imaging using quantitative bioluminescence and single photon imaging was used to measure the distribution of ATP and lactate in two types of human melanoma xenografts with different radiobiologically hypoxic cell fractions (MF: 45 +/- 17% and EE: 6 +/- 3%; mean +/- SD). The tumours were s.c. grown in nude mice and were used for measurement at volumes of 153-3072 mm3. Enhanced chemiluminescent determination of chloramphenicol and related nitro compounds by ‘on-line’ photochemical reaction

The viability of tandem photochemical reaction-chemiluminescence detection was studied for a heterogeneous family of nitro compounds using chloramphenicol as a test substance. The 'on-line' chemical photodegradation of chloramphenicol was performed in a flow injection assembly by using a photoreactor consisting of a 725 cm x 0.5 mm id piece of PTFE tubing coiled around an 8 W low-pressure mercury lamp. Photodegraded chloramphenicol was detected by oxidizing photo-fragments from the parent compound and their subsequent reaction with a luminol-Co(II) system. Isoelectronic series of oxygen deficient centers in silica: experimental estimation of homogeneous and inhomogeneous spectral widths

We report nanosecond time-resolved photoluminescence measurements on the isoelectronic series of oxygen deficient centers in amorphous silica related to silicon, germanium and tin atoms, which are responsible of fluorescence activities at approximately 4 eV under excitation at approximately 5 eV. The dependence of the first moment of their emission band on time and that of the radiative decay lifetime on emission energy are analyzed within a theoretical model able to describe the effects introduced by disorder on the optical properties of the defects. Green phosphorescence and electroluminescence of sulfur pentafluoride-functionalized cationic iridium(III) complexes

We report four cationic iridium(III) complexes [Ir(C^N)2(dtBubpy)](PF6) that have sulfurpentafluoride-modified 1-phenylpyrazole and 2-phenylpyridine cyclometalating (C^N) ligands (dtBubpy = 4,4'-di-tert-butyl-2,2'-bipyridyl). Three of the complexes were characterized by single-crystal X-ray structure analysis. In cyclic voltammetry, the complexes undergo reversible oxidation of iridium(III) and irreversible reduction of the SF5 group. Assessment of toxicity hazards of dredged lake sediment contaminated by creosote.

In order to predict the potential toxicity hazards of sediment remediation by dredging, an experimental laboratory simulation was made by investigating seven ratios of creosote-contaminated sediment (Lake Jamsanvesi, central Finland) and artificial lake water mixtures. Sediment was suspended in water at the ratios of 1:1, 1:2, 1:4, 1:8, 1:16, 1:32, 1:64, 1:128 v/v. The elutriates were analysed for the acute toxicity by photoluminescence bacterial and waterflea (Daphnia magna Straus) tests. The concentrations of polycyclic aromatic hydrocarbons (PAHs) are determined by gas chromatography (GC/FID). Alveolar macrophages (AM) play a decisive role in the immunologic defense system of the lung and in inflammatory pulmonary pathomechanisms. AM and blood monocytes (BM) were exposed to chrysotile B, soot FR 101, and Printex 90 (P 90). We evaluated the reactive oxygen intermediate (ROI) release of AM and BM after particle exposure. ROI release was measured by chemiluminescence. Thirty-minute exposure caused a significant (up to 2.5-fold) increase in ROI release of AM (100 micrograms/10(6) cells) compared with control experiments (p0.01). Differential changes in purine nucleotides after Doxorubicin treatment of human cancer cells in vitro

The present investigation was performed to elucidate the role of purine nucleotides as potential indicators of chemosensitivity of malignant tumors. Drug-sensitive (s) and -resistant (r) tumor cell lines grown as monolayers (s: T47D, MCF-7 wild-type; r: NCI/ADR-RES, MCF-7/MDR) or as multicellular spheroids (T47D; NCI/ADR-RES) were exposed to 0.1, 1.0, and 10.0 microM Doxorubicin for up to 24 h. Chemiluminescence response of ß-glucan stimulated leukocytes isolated from different tissues and peritoneal cavity of Dicentrarchus labrax

Abstract The respiratory burst of leukocytes isolated from sea bass ( Dicentrarchus labrax ) pronephros, peritoneal cavity (P.C.), spleen and blood, was measured by a chemiluminescence (CL) assay after stimulation with β-glucan. The CL response by P.C. and pronephros leukocytes was significantly higher than that expressed by a similar number of cells separated from spleen and blood. This probably reflects the observation that the proportion of macrophages and neutrophils was highest in the populations of leukocytes from peritoneal cavity and pronephros.
