Synthetic multivalent glycopeptide-lipopeptide antitumor vaccines: impact of the cluster effect on the killing of tumor cells.

Multivalent synthetic vaccines were obtained by solid-phase synthesis of tumor-associated MUC1 glycopeptide antigens and their coupling to a Pam3 Cys lipopeptide through click reactions. These vaccines elicited immune responses in mice without the use of any external adjuvant. The vaccine containing four copies of a MUC1 sialyl-TN antigen showed a significant cluster effect. It induced in mice prevailing IgG2a antibodies, which bind to MCF-7 breast tumor cells and initiate the killing of these tumor cells by activation of the complement-dependent cytotoxicity complex.

Abstract Although cancer stem-like cells (CSCs) are rare, they can enter a non-proliferative or dormant state and resist therapy. Furthermore, quiescent CSCs are responsible for metastases that can appear after curative surgical treatment of a primary tumor. Because of drug resistance of CSCs, the development of novel therapies is urgently required that specifically target CSCs. Darba mērķis bija izpētīt pericentriskā heterohromatīna (PHH) iespējamas strukturālas un transkripcijas izmaiņas diferenciācijas sākuma stadijā (komitācijā) ar heregulīnu (HRG) aktivētās MCF-7 krūts vēža šūnās. Modelis tika testēts pēc agrās stresa atbildes gēnu un pericentrisko satelītu DNS transkripcijas dinamikas izmaiņas MCF-7 šūnās pēc HRG aktivācijas izmantojot PĶR. PHH topoloģiju un organizāciju pētīja ar immunofluorescences, konfokālās mikroskopijas un attēlu analīzes metodēm. Tika atklāts, ka PHH klasterizācija ir proporcionāla centromēru skaitam un to izkārtojums raksturojas ar spēka likumsakarību. HRG izsauc PHH strauju de-klasterizāciju. The advances of short interfering RNA (siRNA)-mediated therapy provide a powerful option for the treatment of many diseases, including cancer, by silencing the expression of targeted genes involved in the progression of the pathology. On this regard, a new pH-responsive polycation derived from inulin, Inulin-g-imidazole-g-diethylenetriamine (INU-IMI-DETA), was designed and employed to produce INU-IMI-DETA/siRNA "Inulin COmplex Nanoaggregates" (ICONs). Here, a novel strategy of formulating efficient polymeric carriers based on the already described INU-IMI-DETA for gene material whose structural, functional, and biological properties can be modulated and improved was successfully investigated. In particular, two novel derivatives of INU-IMI-DETA graft copolymer were synthesized by chemical functionalisation with epidermal growth factor (EGF) or polyethylenglycol (PEG), named INU-IMI-DETA-EGF and INU-IMI-DETA-PEG, respectively, in order to improve the performance of already described “inulin complex nanoaggregates” (ICONs).
