Una nuova applicazione della Comparative Multiplex Dosage Analysis (CMDA)

La Comparative Multiplex Dosage Analysis (CMDA) è una tecnica utilizzata per l’analisi di delezioni o duplicazioni di specifiche regioni del genoma (Gable et al. 2003 Hum Mutat 21:379-386). Il metodo si basa su di un’analisi quantitativa delle aree dei picchi di un elettroferogramma ottenuto da elettroforesi capillare condotta su frammenti di DNA amplificati con PCR e marcati con specifici fluorocromi. La presenza di sbilanciamenti è , in particolare, accertata misurando il rapporto tra le aree dei picchi corrispondenti alla regione da analizzare e una regione del genoma la cui dose è nota. Sirt1 è una proteina nota per il suo ruolo di istone deacetilasi NAD+ dipendente che sembra essere coinvolta in una ampia gamma di processi cellulari, quali regolazione genica, controllo dello stato metabolico, meccanismi di sopravvivenza allo stress. Dalla letteratura emergono dati contrastanti concernenti la funzione di Sirt1 nei tumori, le vengono infatti attribuiti ruoli sia di oncogene che di soppressore tumorale, argomento fortemente dibattuto. Sarcophytolide: a new neuroprotective compound from the soft coral Sarcophyton glaucum

Abstract Bioactivity-guided fractionation of an alcohol extract of the soft coral Sarcophyton glaucum collected from the intertidal areas and the fringing coral reefs near Hurghada, Red Sea, Egypt resulted in the isolation of a new lactone cembrane diterpene, sarcophytolide. The structure of this compound was deduced from its spectroscopic data and by comparison of the spectral data with those of known closely related cembrane-type compounds. In antimicrobial assays, the isolated compound exhibited a good activity towards Staphylococcus aureus, Pseudomonas aeruginosa , and Saccharomyces cerevisiae. New complex polycyclic compounds: Synthesis, antiproliferative activity and mechanism of action

Abstract Polycyclic or O-glycoconiugate polycyclic compounds 1a-g were previously tested for their in vitro antiproliferative activity. In this series of compounds, activity increases as log P decreases. Specifically, compounds 1d and 1g showed lower log P values together with the best antiproliferative profiles. With the aim of extending our understanding of the structure–activity relationship (SAR) of this class of compounds, we prepared new polycyclic derivatives 2a-c, which bear on each of the two phenyl rings hydrophilic substituents (OH, SO2NH2 or NHCOCH3). Evaluation of anti-androgenic activity of di-(2-ethylhexyl)phthalate

International audience; DEHP is a widely used platiciser in the manufacture of PVC-based materials. It is known to disrupt the reproductive tract development in male rats. We have performed the Hershberger assay with DEHP on an immature castrated rat model to check if DEHP antagonise the testosterone propionate androgenic effect on the accessory sex organs development. DEHP significantly decreased the BC/LA muscles, the prostate, and the seminal vesicles relative weights from 100, 200, and 400 mg/kg bw/day, respectively.
