Reorganization of myometrial extracellular matrix (ECM) is essential for the uterus to achieve powerful synchronous contractions during labor. Remodeling of the ECM has been implicated in membrane rupture and cervical ripening. Because maternal obesity is associated with both delivery disorders and elevated circulating leptin levels, this study aimed to assess the ability of leptin to interfere with lipopolysaccharide (LPS)-induced myometrial ECM remodeling. In recent years, it has been demonstrated that Hsp70 is released in the extracellular space under normal cell culture conditions, and this release is mediated through exosomes. We have demonstrated that A6 cells, a clone of mouse mesoangioblasts, produce and release in the extracellular space membrane vesicles, independently of culture growth conditions. These vesicles contains both structural proteins and biological active molecules, such as FGF-2 and the metalloproteinases MMP 2 and 9. The human gene Hugl-1 (Llgl/Lgl1) has significant homology to the Drosophila tumor suppressor gene lethal(2)giant larvae (lgl). The lgl gene codes for a cortical cytoskeleton protein, Lgl, that is involved in maintaining cell polarity and epithelial integrity.
