Keyhole limpet hemocyanin (KLH), II: Characteristic reassociation properties of purified KLH1 and KLH2.
1997
Subunits of the two types of keyhole limpet hemocyanin (KLH1 and KLH2), purified by gel filtration chromatography and preparative polyacrylamide gel electrophoresis from Immucothel, have been used for macromolecular reassociation studies. In-vitro reassociation has been achieved with a standardized system using a Tris-saline stabilizing buffer at pH 7.4 containing 100 mM calcium and magnesium chloride at 4 degrees C. The relatively slow progress of reassociation has been monitored and the varying oligomeric forms of KLH1 and KLH2 produced have been studied by transmission electron microscopy, using specimens negatively stained with 5% ammonium molybdate containing 1% trehalose. Specimens ha…
8-Azido-adenosine 5'-triphosphate as a Photoaffinity Label for Bacterial F1 ATPase
1978
1. 8-Azido-adenosine 5'-triphosphate (n83ATP) is a suitable photoaffinity label for F1 ATPase from Micrococcus luteus. The nucleotide is a substrate in the presence of bivalent cations and inhibits the enzyme irreversibly upon irradiation with ultraviolet light above 300 nm. 2. More than 80% of the label is covalently bound to the beta subunits in the presence of bivalent cations. Labeling and inactivation is decreased by protection with ADP, ATP or adenyl-5'-yl imidodiphosphate. To a much smaller degree the alpha subunits also become labeled. 3. n83AMP does not specifically bind to the beta subunits upon irradiation. Like n83ATP and n83ADP, it also labels the alpha subunits to a small exte…
High purity recovery of magnesium and calcium hydroxides from waste brines
2021
Direct disposal of concentrate brines, generated by many industrial sectors, causes serious environmental concerns. An industrial sector that produces high concentrate brines is softening water industry, in particular during the regeneration of ion exchange resins. These brines are mainly rich in sodium, magnesium and calcium chlorides, thus, their regeneration, by removal/recovery of magnesium and calcium, can be a valuable option to turn the brine into a source of minerals and reduce disposal costs and environmental concerns. In this regards, one of the goals of EU-funded ZERO BRINE project is to develop a treatment chain for the valorization and regeneration of spent brines from water so…
Experimental and numerical analysis on post welding formability of FSWed AZ31 magnesium alloy thin joints obtained using a "Pinless" tool configurati…
2012
The post welding formability of friction stir welded AZ31 magnesium alloy thin sheets (1.5 mm thick), obtained using a “pinless” tool configuration, was widely investigated by means of the hemispherical punch method at 350°C, with a constant crosshead speed of 0.1 mm/s. The results were compared with those obtained on the base material. It has shown that formability of the joints is lower than the one of the base material. The experimental work was supported by a numerical investigation based on FEM in order to highlight the material flow occurring during the welding process. Additionally, hemispherical punch tests were simulated starting from the calculated conditions, in terms of accumula…

Power monitoring in dielectric-loaded surface plasmon-polariton waveguides
2011
We report on propagating mode power monitoring in dielectric-loaded surface plasmon-polariton waveguides (DLSPPWs) by measuring the resistance of gold stripes supporting the DLSPPW mode propagation. Inevitable absorption of the DLSPPW mode in metal causes an increase in the stripe temperature and, thereby, in its resistance whose variations are monitored with an external Wheatstone bridge being accurately balanced in the absence of radiation in a waveguide. The investigated waveguide configuration consists of a 1-µm-thick and 10-µm-wide polymer ridges tapered laterally to a 1-µm-wide ridge placed on a 50-nm-thin and 4-µm-wide gold stripe, all supported by a magnesium fluoride substrate. Usi…
Fiber-coupled dielectric-loaded plasmonic waveguides.
2010
Fiber in- and out-coupling of radiation guided by dielectric-loaded surface plasmon-polariton waveguides (DLSPPWs) is realized using intermediate tapered dielectric waveguides. The waveguide structures fabricated by large-scale UV-lithography consist of 1-microm-thick polymer ridges tapered from 10-microm-wide ridges deposited directly on a magnesium fluoride substrate to 1-microm-wide ridges placed on a 50-nm-thick and 100-microm-wide gold stripe. Using fiber-to-fiber transmission measurements at telecom wavelengths, the performance of straight and bent DLSPPWs is characterized demonstrating the overall insertion loss below 24 dB, half of which is attributed to the DLSPPW loss of propagati…
