Effects of indole-3-acetic acid on Sinorhizobium meliloti survival and on symbiotic nitrogen fixation and stem dry weight production

2009

We evaluated the effects of the main auxin phytohormone, indole-3-acetic acid (IAA), on the central metabolism of Sinorhizobium meliloti strain 1021. We either treated the Sinorhizobium meliloti 1021 strain with 0.5 mM IAA (1021+) or use a derivative, RD64, of the same strain harbouring a pathway for IAA biosynthesis converting tryptophan into IAA via indoleacetamide. We assayed the activity of key enzymes in the major energy-yielding pathways (Entner-Doudoroff, Embden-Meyerhof-Parnas, pentose phosphate, glyoxylate bypass and tricarboxylic acid cycle). We found that activity of two main regulative tricarboxylic acid (TCA) cycle enzymes was increased. Citrate synthase (CS) activity, as compa…

Preliminary insights into the molecular barcoding data of Turraea socotrana (Meliaceae) from Socotra (Yemen)

2020

The relationship of two geographically and ecologically separated populations of Turraea socotrana Styles and F. White (Meliaceae), a vulnerable and scattered shrub from the island of Socotra (Yemen), was explored using preliminary molecular data. The nuclear ribosomal ITS region was used to screen the level of genetic divergence of T. socotrana from ecologically distant localities in comparison to a few other species of Turraea and closely related genera. The individuals from the inland (Diksam) differed from the western coastal ones (Ma’alah) by vegetative characters and by the rbcL gene. This may be due to the fact that the population from Ma’alah grows on lithosoil slopes directly expos…

Proteome analysis and identification of symbiosis-related proteins from Medicago truncatula Gaertn. by two-dimensional electrophoresis and mass spect…

2002

Time-course analysis of root protein profiles was studied by two-dimensional gel electrophoresis and silver staining in the model plant Medicago truncatula, inoculated either with the arbuscular mycorrhizal fungus Glomus mosseae or with the nitrogen fixing bacterium Sinorhizobium meliloti. Protein modifications in relation to the development of both symbioses included down- and upregulations, as well as newly induced polypeptides. Matrix assisted laser desorption/ionization-time of flight-mass spectrometry after trypsin digestion clearly identified one polypeptide induced in nodulated roots as a M. truncatula leghemoglobin. Internal sequencing with a quadrupole time-of-flight mass spectrome…

Anthropic space and design for all. New knowledge paths for urban planning strategies

2019

Abstract Nowadays city environment shows the presence of a mixed variety of elements, as natural, semi natural and anthropic components that build up both structure and connections of the urban context. This specific structure shapes and directs space and its functions strictly connected with their sustainable potential uses and sustainable development opportunities. The lack of rules and proper planning methods produces inefficient use conditions by resident citizens, entropy, functions' reduction of ecological networks and deep environmental impacts. The consequence comes out to be a great widespread life quality decrease in urban areas. These thoughts lead the authors to rethink the defi…

Quantitative methods in tourism and hospitality: a perspective article

2019

Purpose The purpose of this paper is to provide an overview of past perspectives and future trends in tourism and hospitality research. Design/methodology/approach The study grounds the discussion on the timeline evolution of quantitative research methods. Findings Although still under-recognized by scholars, mixed methods represent the future of research in tourism and hospitality. Research limitations/implications The investigation is confined to quantitative methods. Originality/value No other surveys sketch a period of 150 years of quantitative analyses in tourism and hospitality.

High-performance liquid chromatographic separation of modified and native melittin following transglutaminase-mediated derivatization with a dansyl f…

1991

Abstract The 26-amino acid linear, amphiphilic peptide melittin was enzymatically modified with the fluorescent probe monodansylcadaverine using guinea pig liver transglutaminase and a fluorescent derivative of stoichiometry 1:1 was obtained. Reversed-phase and size-exclusion high-performance liquid chromatographic modes were tested in order to resolve the labelled peptide and native species. The influence of several operational variables was analysed and the elution conditions were optimized so that a satisfactory resolution could be achieved in both instances in a rapid, easy manner. Both chromatographic modes offer the possibility of accurate monitoring of the time course of the enzyme-m…
