An experimental study of the reproductive success of Echinostoma friedi (Trematoda: Echinostomatidae) in the golden hamster

Viable eggs produced weekly per infective stage was used as a measure of the reproductive success of Echinostoma friedi during the first 12 weeks of infection in hamsters. The weekly reproductive success was not constant during the experiment in relation to the egg output and the proportion of viable eggs produced. The egg release started during week 2 post-inoculation, attaining a maximum during week 3. A decline in egg output was observed from week 9. Inforimation on the ambient lighting conditions is conveyed from the retina to the pineal organ by a neuronal pathway involving the suprachiasmatic nucleus (SCN) which acts as a circadian pacemaker. In the hamster, circadian rhythms have been shown to be influenced by injection of neuropeptide Y (NPY) into the SCN. Since NPY-immunoreactive nerve fibres are present in the rat and guinea-pig pineal glands it appeared of interest to investigate the hamster pineal as part of the circadian rhythm generating/regulating system. In cremaster arterioles, the contribution of a myogenic component to the constriction on intravenous infusion of norepinephrine (NE) or angiotensin II (Ang II) was studied. Elevation of plasma viscosity induces sustained NO-mediated dilation in the hamster cremaster microcirculation in vivo

We studied whether a flow-independent increase of luminal wall shear stress (WSS) could dilate hamster arterioles in vivo and which endothelial mediators are potentially involved. To this end the plasma viscosity was elevated by exchanging blood for dextran-erythrocyte solution thereby augmenting WSS. Studies on the disposition, metabolism and hepatotoxicity of coumarin in the rat and Syrian hamster.

The hepatotoxicity, metabolism and disposition of coumarin has been compared in male Sprague-Dawley rats and Syrian hamsters. The treatment of rats for 12, 24 and 42 weeks with diets containing 0.2 and 0.5% coumarin resulted in hepatotoxicity and increased relative liver weights. While levels of cytochrome P450 (CYP) and CYP-dependent enzymes were decreased, levels of reduced glutathione (GSH) and activities of UDP glucuronosyltransferase, gamma-glutamyltransferase and GSH S-transferase were increased. Electrophysiological characterization of the pineal gland of golden hamsters.

In one of the most-widely used species in pineal gland research, the strongly photoperiodic golden hamster, Mesocricetus auratus, no electrophysiological data on pinealocytes are currently available. To fill this gap, in the present study 185 spontaneously active pinealocytes from male golden hamsters were recorded extracellularly, both during day- and night-time (light: dark cycle 12 ∶ 12, lights on at 07.00 h). As in other species, pinealocytes exhibited action potentials of 1–2 ms duration. An irregular firing pattern was observed in 95% of the pinealocytes, the remainder fired more regularly or showed a phasic discharge pattern. Nalidixic acid-resistant V79 cells with reduced DNA topoisomerase II activity and amplification prone phenotype

Spontaneously nalidixic acid-resistant lines (NAr lines) were selected from a V79 Chinese hamster cell line and phenotypically characterized. NAr lines showed an increased doubling time, a higher number of spontaneous SCE, and more interestingly, decreased DNA topoisomerase II activity. These lines were also cross-resistant to the eukaryotic topoisomerase II inhibitors etoposide and adriamycin, but showed the same level of sensitivity as the parental line to the DNA topoisomerase I inhibitor camptothecin. An in vitro model to study cellular photosensitizer uptake and photodynamic dose-response relationships of tumor cells

Cellular fluorescence intensity (CFI) after incubation with varying concentrations of the photosensitizer Photofrin and the photodynamically induced dose-response relationships of hamster melanoma cells (A-MEL-3) were studied in a recently developed in vitro model. After administration of Photofrin to the extracellular serum-free medium, CFI was evaluated by flow cytometry together with constantly fluorescing latex particles used as a reference. Glutathione content of V79 cells in two- or three-dimensional culture

The cellular glutathione (GSH) content of two- and three-dimensional cell cultures of V79 hamster lung cells has been studied. As previously described, cells in monolayer cultures show a decrease in GSH when they reach the confluent state. Three-dimensional cell cultures (multicell spheroids) allow a smoother transition from the initial proliferating to the nonproliferating status, and they show a central area of necrosis when a certain diameter is reached.
