The metabolic responses to electrical pulse-induced contractions in C2C12 myotubes are greater in high glucose medium
2021
Osaa liikunnan terveyshyötyjen mekanismeista on vaikeaa tutkia ihmisillä tai koe-eläimillä. Viljeltyjen C2C12 myotuubien sähköpulssistimulaatiolla (EPS, engl. electrical pulse stimulation) voidaan mallintaa lihassupistuksia ja liikuntaa in vitro –olosuhteissa.
Untargeted metabolomics of rind essential oils allowed to differentiate two closely related clementine varieties
2021
[EN] Chemical characterization of clementine varieties (Citrus clementina Hort. ex Tan.) essential oils (EO) can lead to variety identification and valorization of their potential use in food and aroma industries. The goal of this study was the chemometric discrimination between two very closely related and morphologically identical clementine varieties, Clemenules (NL) and Clemenpons (PO), based on their rind EO, to identify the differential volatile organic compounds (VOCs) and to determine their antioxidant capacity. EO rind volatile profile was determined by gas chromatography coupled to mass spectrometry in Citrus fruit at different ripening stages grown two independent years in two di…
Mixotrophy in a Local Strain of Nannochloropsis granulata for Renewable High-Value Biomass Production on the West Coast of Sweden
2022
A local strain of Nannochloropsis granulata (Ng) has been reported as the most productive microalgal strain in terms of both biomass yield and lipid content when cultivated in photobioreactors that simulate the light and temperature conditions during the summer on the west coast of Sweden. To further increase the biomass and the biotechnological potential of this strain in these conditions, mixotrophic growth (i.e., the simultaneous use of photosynthesis and respiration) with glycerol as an external carbon source was investigated in this study and compared with phototrophic growth that made use of air enriched with 1–2% CO2. The addition of either glycerol or CO2-enriched air stimulated the…
1H-NMR-Based Metabolomics in Autism Spectrum Disorder and Pediatric Acute-Onset Neuropsychiatric Syndrome
2022
We recently described a unique plasma metabolite profile in subjects with pediatric acute-onset neuropsychiatric syndrome (PANS), suggesting pathogenic models involving specific patterns of neurotransmission, neuroinflammation, and oxidative stress. Here, we extend the analysis to a group of patients with autism spectrum disorder (ASD), as a consensus has recently emerged around its immune-mediated pathophysiology with a widespread involvement of brain networks. This observational case-control study enrolled patients referred for PANS and ASD from June 2019 to May 2020, as well as neurotypical age and gender-matched control subjects. Thirty-four PANS outpatients, fifteen ASD outpatients, an…
Promoter Activation in Dhfq Mutants as an Efficient Tool for Specialized Metabolite Production Enabling Direct Bioactivity Testing
2019
Abstract Natural products (NPs) from microorganisms have been important sources for discovering new therapeutic and chemical entities. While their corresponding biosynthetic gene clusters (BGCs) can be easily identified by gene‐sequence‐similarity‐based bioinformatics strategies, the actual access to these NPs for structure elucidation and bioactivity testing remains difficult. Deletion of the gene encoding the RNA chaperone, Hfq, results in strains losing the production of most NPs. By exchanging the native promoter of a desired BGC against an inducible promoter in Δhfq mutants, almost exclusive production of the corresponding NP from the targeted BGC in Photorhabdus, Xenorhabdus and Pseud…

Potential of ‘Omics’ Technologies for Implementation in Research on Phytotherapeutical Toxicology
2012
Abstract High toxicity is the most common reason why new agents drop out of drug development in the pharmaceutical industry. There is hope that toxicogenomics facilitates the early detection of toxic effects and their molecular mechanisms of action during preclinical studies to remove potentially toxic substances from the development. Herbal remedies consist of mixtures of different herbs, which represent a considerable source of heterogeneity and toxicity. They may be caused by botanical misidentification, contamination with pesticides, heavy metals, organic solvents, microbials and radioactivity. Intentional faked herbal products may contain chemical drugs or hormones. Approaches to apply…
Tear 1H Nuclear Magnetic Resonance-Based Metabolomics Application to the Molecular Diagnosis of Aqueous Tear Deficiency and Me…
2020
Purpose: Meibomian gland dysfunction (MGD) is a major cause of signs and symptoms related to dry eyes (DE) and eyelid inflammation. We investigated the composition of human tears by metabolomic approaches in patients with aqueous tear deficiency and MGD. Methods: Participants in this prospective, case-control pilot study were split into patients with aqueous tear deficiency and MGD (DE-MGD [n = 15]) and healthy controls (CG; n = 20). Personal interviews, ocular surface disease index (OSDI), and ophthalmic examinations were performed. Reflex tears collected by capillarity were pr…
