Differential staining of peripheral nuclear chromatin with Acridine orange implies an A-form epichromatin conformation of the DNA

ABSTRACT The chromatin observed by conventional electron microscopy under the nuclear envelope constitutes a single layer of dense 30–35 nm granules, while ∼30 nm fibrils laterally attached to them, form large patches of lamin-associated domains (LADs). This particular surface “epichromatin” can be discerned by specific (H2A+H2B+DNA) conformational antibody at the inner nuclear envelope and around mitotic chromosomes. A novel method for determination of inorganic polyphosphates using the fluorescent dye fura-2.

A method for determining inorganic polyphosphate, which is based on the Mn2+-induced quenching of the fluorescence of the calcium indicator fura-2, is described. The effect of Mn2+ ions on fura-2 fluorescence is gradually abolished in the presence of increasing concentrations of polyphosphate; this allows the quantification both of synthetic polyphosphates and of the naturally occurring polymer isolated from tissues or cells. The described method has some advantages compared to conventional procedures for detection of polyphosphates based on the metachromatic effect on toluidine blue. Metachromatic staining and electron dense reaction of glycosaminoglycans by means of Cuprolinic Blue

The cationic phthalocyanin-like dye Cuprolinic Blue, unlike phthalocyanin dyes such as Alcian Blue or Astra Blue, can definitely exhibit a clear metachromatic reaction with appropriate substrates. The application of Cuprolinic Blue to epoxy-embedded semithin sections revealed that mast cell cytoplasmic granules, goblet cell mucin and cartilage matrix stained in violet shades (metachromatic), whereas nuclear chromatin presented a bright blue coloration (orthochromatic). Post-synovectomy changes in the articular cartilage

Synovectomy of the left knee was performed in 37 immature rabbits, using the right knee for control. In the first set of experiments the articular cartilage was examined at weekly intervals for 8 weeks following the operation, paying particular attention to the metachromatic changes in the ground substance of the cartilage. In the second set of experiments, restoration of synovium was examined. In the third and fourth set of experiments, the uptake of S35 by the cartilage was assessed using autoradiography and densitometry. In the fifth set of experiments, alterations in S35 uptake by the chondrocitic cells of the cartilage were studied by electron microscopy.
