Effect of ethanol on yeast film formation

1999

<p style="text-align: justify;">In this study, we have investigated the influence of ethanol on yeast film formation and cell surface hydrophobicity (CSH). A yeast strain (P3) previously isolated from film yeast was grown in a medium containing increasing ethanol concentration ranging from 0 to 14 p. cent (v/v). It results from this study that up to 10 p. cent ethanol, the greater was the ethanol concentration, the greater was the growth of film. Using two different techniques (phase partition method, magnobead assay), we have shown that ethanol altered the CSH of the yeast. The measured hydrophobicity (p. cent) of cells grown without ethanol was 65 p. cent compared with 81 p. cent wi…

Experimental and fluid dynamic study of continuous supercritical water gasification of glucose

2016

Abstract The supercritical water gasification (SCWG) of glucose as a model compound for H 2 production is studied in term of process development at 25 MPa and 650 °C. Gasification has been investigated using a laboratory scale continuous plant comprising a continuous down flow reactor with a volume of 200 cm 3 , a heat recovery section and pre-heat of the biomass to simulate more closely an industrial design. Feed streams (pure water and glucose solution) are introduced at the top of the vessel and products are extracted from the bottom. The reactor is equipped with various inlet points that allow to modify the inlet position of reacting streams in order to study the effect of stream mixing…

Voltammetric determination of dipeptidyl aminopeptidases I and IV

1992

Dipeptidyl aminopeptidases I and IV have recently found wide practical application (polypeptide sequencing, clinical diagnostics) so it is necessary to extend present methods and work out new ones for determination of their activity. A new, voltammetric method for determination of peptidase activity is proposed, which allows optimization of the determination conditions according to the activity to be measured. The method has some advantages over the spectrophotometric methods, such as applicability to opaque solutions and higher sensitivity.

A microscale Kjeldahl nitrogen determination for environmental waters.

2008

A microscale Kjeldahl system has been designed which has been homemade reduced. The digestor unit of a macroKjeldahl system and a modified glassware microdistillation unit have been used. The optimal conditions for digestion and distillation have been established for ammonium and methylamine as model compound of organic amino nitrogen. The optimised procedure has been applied to the determination of Kjeldahl nitrogen in water samples. Recoveries near 100% and good precision have been achieved. This procedure combines nitrogen miniaturized system and the use of a selective ion electrode for ammonia detection. The analysis time was reduced 4.8 times and the analysis cost 6.6 times compared wi…

Robust interpretive optimisation in high-performance liquid chromatography considering uncertainties in peak position.

2005

In the context of interpretive chromatographic optimisation, robustness is usually calculated by introducing deliberated shifts in the nominal optimal conditions and evaluating their effects on the monitored objective function, mimicking thus the experimental procedures used in method validation. However, such strategy ignores a major source of error: the uncertainties associated to the modelling step, that may give rise to deceiving results when conditions that were expected to yield baseline separation are reproduced in the chromatograph. Two approaches, based on the peak purity concept, are here proposed to evaluate the robustness of the objective function under the perspective of measur…

Use of protein profiles established by CZE to predict the cultivar of olive leaves and pulps

2014

Intact protein profiles established by CZE have been used to predict the cultivar of olive leaves and pulps. For this purpose, proteins were extracted using a mild enzyme-assisted extraction method, which provided higher protein recoveries and a lower environmental impact than other previously described methods. These extracts were subjected to CZE determination under basic conditions using a BGE composed of 50 mM phosphate, 50 mM tetraborate and 0.1% PVA at pH 9. Nine and 14 common peaks, for leaf and pulp samples, respectively, were identified in the nine cultivars studied in this work. In addition, and using linear discriminant analysis of the CZE data, olive leaf and pulp samples belong…

Retrospective screening of pesticide metabolites in ambient air using liquid chromatography coupled to high-resolution mass spectrometry

2015

A new methodology for the retrospective screening of pesticide metabolites in ambient air was developed, using liquid chromatography coupled to Orbitrap high-resolution mass spectrometry (UHPLC-HRMS), including two systematic workflows (i) post-run target screening (suspect screening) and (ii) non-target screening. An accurate-mass database was built and used for the post-run screening analysis. The database contained 240 pesticide metabolites found in different matrixes such as air, soil, water, plants, animals and humans. For non-target analysis, a "fragmentation-degradation" relationship strategy was selected. The proposed methodology was applied to 31 air samples (PM10) collected in the…

Analytical Methods for Pesticide Residue Determination in Bee Products

2002

Monitoring pesticide residues in honey, wax, and bees helps to assess the potential risk of these products to consumer health and gives information on the pesticide treatments that have been used on the field crops surrounding the hives. The present review seeks to discuss the basic principles and recent developments in pesticide analysis in bee products and their application in monitoring programs. Consideration is given to extraction, cleanup, chromatographic separation, and detection techniques.

Infrared biospectroscopy for a fast qualitative evaluation of sample preparation in metabolomics.

2014

Liquid chromatography-mass spectrometry (LC-MS) has been increasingly used in biomedicine to study the dynamic metabolomic responses of biological systems under different physiological or pathological conditions. To obtain an integrated snapshot of the system, metabolomic methods in biomedicine typically analyze biofluids (e.g. plasma) that require clean-up before being injected into LC-MS systems. However, high resolution LC-MS is costly in terms of resources required for sample and data analysis and care must be taken to prevent chemical (e.g. ion suppression) or statistical artifacts. Because of that, the effect of sample preparation on the metabolomic profile during metabolomic method d…
