Abstract Purpose: 3-hydroxy-3-methylglutaryl CoA reductase inhibitors (statins) are frequently used lipid-lowering drugs. Moreover, they are reported to exert pleiotropic effects on cellular stress responses, proliferation, and apoptosis. Temozolomide- and fotemustine-induced apoptosis in human malignant melanoma cells: response related to MGMT, MMR, DSBs, and p53

Malignant melanomas are highly resistant to chemotherapy. First-line chemotherapeutics used in melanoma therapy are the methylating agents dacarbazine (DTIC) and temozolomide (TMZ) and the chloroethylating agents BCNU and fotemustine. Here, we determined the mode of cell death in 11 melanoma cell lines upon exposure to TMZ and fotemustine. We show for the first time that TMZ induces apoptosis in melanoma cells, using therapeutic doses. For both TMZ and fotemustine apoptosis is the dominant mode of cell death. Topotecan-triggered degradation of topoisomerase I is p53-dependent and impacts cell survival.

Abstract The anticancer drug topotecan belongs to the group of topoisomerase I (topo I) inhibitors. In the presence of topotecan, topo I cleaves the DNA but is unable to religate the single-strand break. This leads to stabilization of topo I-DNA–bound complexes and the accumulation of DNA strand breaks that may interfere with DNA replication. The molecular mechanism of controlling the repair of topo I-DNA covalent complexes and its impact on sensitivity of cells to topotecan is largely unknown. Uniformization with infinitesimally metric measures

We consider extensions of quasiconformal maps and the uniformization theorem to the setting of metric spaces $X$ homeomorphic to $\mathbb R^2$. Given a measure $\mu$ on such a space, we introduce $\mu$-quasiconformal maps $f:X \to \mathbb R^2$, whose definition involves deforming lengths of curves by $\mu$. We show that if $\mu$ is an infinitesimally metric measure, i.e., it satisfies an infinitesimal version of the metric doubling measure condition of David and Semmes, then such a $\mu$-quasiconformal map exists. We apply this result to give a characterization of the metric spaces admitting an infinitesimally quasisymmetric parametrization.

Modelling of thermo-chemical properties over the sub-solidus MgO–FeO binary, as a function of iron spin configuration, composition and temperature

Thermo-chemical properties and T–X phase relations diagram of the (Mg,Fe)O solid solution are modelled using mixing Helmholtz energy, ΔF(T,x)mixing, calculated by quantum mechanical and semi-empirical techniques. The sub-solidus MgO–FeO binary has been explored as a function of composition, with iron either in high-spin (HS) or low-spin (LS) configuration. Only the HS model provides physically sound results at room pressure, yielding a correct trend of cell edge versus composition, whereas LS’s issues are at variance with observations. The HMG-CoA reductase inhibitor lovastatin protects cells from the antineoplastic drugs doxorubicin and etoposide

Ras-homologous GTPases are involved in the regulation of genotoxic stress-induced gene expression and cell death. Since they need C-terminal isoprenylation for correct intracellular localization and function, we investigated whether depletion of cells from isopren precursor moieties using the HMG-CoA reductase inhibitor lovastatin affects cellular sensitivity to DNA damaging drugs. Here we show that lovastatin renders cells highly resistant to the tumor-therapeutic compound doxorubicin. Desensitization by lovastatin was reverted by co-treatment with GGPP indicating that inhibition of protein geranylgeranylation is involved in acquired doxorubicin resistance. Local minimizers and gamma-convergence for nonlocal perimeters in Carnot groups

We prove the local minimality of halfspaces in Carnot groups for a class of nonlocal functionals usually addressed as nonlocal perimeters. Moreover, in a class of Carnot groups in which the De Giorgi's rectifiability Theorem holds, we provide a lower bound for the $\Gamma$-liminf of the rescaled energy in terms of the horizontal perimeter.

Semmes surfaces and intrinsic Lipschitz graphs in the Heisenberg group

A Semmes surface in the Heisenberg group is a closed set $S$ that is upper Ahlfors-regular with codimension one and satisfies the following condition, referred to as Condition B. Every ball $B(x,r)$ with $x \in S$ and $0 < r < \operatorname{diam} S$ contains two balls with radii comparable to $r$ which are contained in different connected components of the complement of $S$. Analogous sets in Euclidean spaces were introduced by Semmes in the late $80$'s.
