Small angle scattering study of poly(methylmethacrylate)-block-poly(ethylene oxide) block co-polymer in aqueous solution
2005
A combined Small Angle X ray (SAXS) and Neutron (SANS) Scattering study of aqueous solutions of a symmetric block copolymer consisting of poly(methyl methacrylate) (PMMA) and poly(ethylene oxide) (PEO) moieties is presented. The polymer forms slightly polydisperse spherical micelles in a wide range of concentration (0.03 – 6.7 w/V) and temperature (20°C ≤ T ≤ 65°C). A good description of the SANS data is obtained using a polydisperse core-shell model with a structure factor for a modified hard sphere potential. By increasing the concentration at constant T we observed a decrease of the aggregation number and an increase of solvation of PEO groups in the shell, opposite to what happens by in…
Novel inulin-based mucoadhesive micelles loaded with corticosteroids as potential transcorneal permeation enhancers
2017
In this work a new copolymer of inulin (INU) derivatized with ethylendiamine (EDA) and retinoic acid (RA), named INU-EDA-RA, was synthetized, characterized and employed to produce micelles as carriers for topical administration of corticosteroids for the potential treatment of diseases of posterior eye segment. Spectroscopic analysis confirmed a molar derivatization degree of 11.30 and 4.30% in EDA and RA, respectively. INU-EDA-RA micelles are capable of strong mucoadhesive interactions which result time-independent and stable over time but concentration depending. Moreover micelles are able to encapsulate efficiently from 3 to 13% (w/w) of lipophilic drugs, as dexamethasone, triamcinolone …
Biopartitioning micellar separation methods: modelling drug absorption
2003
The search for new pharmacologically active compounds in drug discovery programmes often neglects biopharmaceutical properties as drug absorption. As a result, poor biopharmaceutical characteristics constitute a major reason for the low success rate for candidates in clinical development. Since the cost of drug development is many times larger than the cost of drug discovery, predictive methodologies aiding the selection of bioavailable drug candidates are of profound significance. This paper has been focussed on recent developments and applications of chromatographic systems, particularly those systems based on amphiphilic structures, in the frame of alternative approaches for estimating t…
Ruthenium-Containing Block Copolymer Assemblies: Red-Light-Responsive Metallopolymers with Tunable Nanostructures for Enhanced Cellular Uptake and An…
2015
The use of self-assembled nanostructures consisting of red-light-responsive Ru(II)-containing block copolymers (BCPs) for anticancer phototherapy is demonstrated. Three Ru-containing BCPs with different molecular weights are synthesized. Each BCP contains a hydrophilic poly(ethylene glycol) block and an Ru-containing block. In the Ru-containing block, more than half of the side chains are coordinated with [Ru(2,2':6',2''-terpyridine)(2,2'-biquinoline)](2+) , resulting in more than 40 wt% Ru complex in the BCPs. The Ru complex acts as both a red-light-cleavable moiety and a photoactivated prodrug. Depending on their molecular weights, the BCPs assemble into micelles, vesicles, and large comp…
Retention mechanisms for basic drugs in the submicellar and micellar reversed-phase liquid chromatographic modes.
2008
The reversed-phase liquid chromatographic (RPLC) behavior (retention, elution strength, selectivity, efficiency, and peak asymmetry) for a group of basic drugs (beta-blockers), with mobile phases containing the anionic surfactant sodium dodecyl sulfate (SDS) and acetonitrile, revealed different separation environments, depending on the concentrations of both modifiers: hydro-organic, submicellar at low surfactant concentration and high concentration of organic solvent, micellar, and submicellar at high concentration of both surfactant and organic solvent. In the surfactant-mediated modes, the anionic surfactant layer adsorbed on the stationary phase interacts strongly with the positively ch…
Investigations of methyl lactate in the presence of reverse micelles by vibrational spectroscopy and circular dichroism
2012
Abstract The FT-IR and vibrational circular dichroism (VCD) spectra of ( S )- and -( R )-methyl lactate have been recorded for neat samples and at various concentrations in CCl 4 and DMSO solutions. These spectra are used to analyse the FT-IR and VCD spectra of methyl lactates in presence of sodium bis(2-ethylhexyl)sulfosuccinate (AOT) in CCl 4 , where the surfactant molecules are known to form reverse micelles. Some tendency of methyl lactate to interact with AOT micellar aggregates is observed, but not as well defined as previously observed for dimethyl tartrate in analogous circumstances. Besides, near infrared (NIR) absorption and VCD data have been obtained for most of the above system…
About entangled networks of worm-like micelles: a rejected hypothesis
1996
We report new results from small-angle neutron scattering on d(1 2)-cyclohexane/lecithin/water micellar solutions performed as a function of the water content (w(o)), temperature (T) and dispersed phase volume fraction (phi). The data from dilute samples are interpretable in terms of the existence of giant cylindrical reverse micelles and are well fit with a core-shell model (that provides the micelle structure and dimensions) with values of 28 and 45 Angstrom for the inner core and the outer shell radii, almost independent on temperature and concentration. Such a result could appear consistent with the current idea that worm-like micelles are living polymers. On the contrary, the appearanc…
Studies of the physicochemical and structural properties of self-assembling cationic pyridine derivatives as gene delivery agents.
2015
New amphiphilic pyridine derivatives containing dodecyloxycarbonyl substituents at positions 3 and 5 and cationic moieties at positions 2 and 6 have been designed and synthesised. Compounds of this type can be considered as synthetic lipids. The corresponding 1,4-dihydropyridine (1,4-DHP) derivatives have earlier been proposed as a promising tool for plasmid DNA (pDNA) delivery in vitro. In this work studies of the self-assembling properties of amphiphilic pyridine derivatives leading to the formation of liposomes, determination of particle size, zeta-potential and critical micelle concentration (CMC) with dynamic light scattering (DLS) measurements are described. Furthermore, thermal analy…
Fluorinated and pegylated polyaspartamide derivatives to increase solubility and efficacy of Flutamide
2012
New fluorinated amphiphilic copolymers based on a biocompatible polyaspartamide have been prepared in order to obtain polymeric micelles useful for delivering anticancer drugs. In particular, α,β-poly(N-2-hydroxyethyl)-d,l-aspartamide (PHEA) has been derivatized with polyethylene glycol (PEG(2000)) and ethylendiamine (EDA). Both these portions form the hydrophilic part of the copolymer, while the hydrophobic moiety is given by 1,2,4-oxadiazoles: 5-pentafluorophenyl-3-perfluoroheptyl-1,2,4-oxadiazole (PPOX) or 3-carboxyethyl-5-pentadecafluoroheptyl-1,2,4-oxadiazole (CPOX). Copolymers named PHEA-PEG(2000)-EDA-PPOX and PHEA-PEG(2000)-EDA-CPOX have been prepared with various degrees of derivati…
Physicochemical investigation of acrylamide solubilization in sodium bis(2-ethylhexyl)sulfosuccinate and lecithin reversed micelles.
2003
The state of acrylamide confined within dry sodium bis(2-ethylhexyl)sulfosuccinate (AOT) and lecithin reversed micelles dispersed in CCl4 has been investigated by FTIR and H-1 NMR spectroscopy. Measurements have been performed at 25 degreesC as a function of the acrylamide-to-surfactant molar ratio (R) at a fixed surfactant concentration (0.1 mol kg(-1)). The analysis of experimental data, corroborated by the results of SAXS measurements, is consistent with the hypothesis that acrylamide is quite uniformly distributed among reversed micelles mainly located in proximity to the surfactant head-group region and that its presence induces significant unidimensional growth of micellar aggregates.…
