Inactivation of peroxisomal ABC transporters, ABCD1 and ABCD2 in BV-2 microglial cells : Towards a better understanding of X-linked adrenoleukodystro…
2018
X-linked adrenoleukodystrophy (X-ALD) is a severe neurodegenerative disorder characterized by very-long-chain fatty acid (VLCFA) accumulation resulting from a peroxisomal β-oxidation defect. The disease is caused by mutations in the ABCD1 gene, which encodes for a peroxisomal half ABC transporter predicted, like its closest homologue ABCD2, to participate in the entry of VLCFA-CoA into the peroxisome, the unique site of their β-oxidation. Progress in understanding the physiopathogenesis of X-ALD suffers from the lack of appropriate cell and animal models. Since peroxisomal defects in microglia seem to be a key element of the onset of the disease, we generated four microglial cell lines unab…
Effets d’extraits de raquette du cactus -Opuntia ficus indica- sur la modulation du stress oxydant et du processus inflammatoire liés à la déficienc…
2018
Acyl-CoA oxidase 1 (ACOX1) deficiency is a rare and severe peroxisomal leukodystrophy associated with a very long-chain fatty acids β-oxidation defect. As peroxisomal defects in microglia appear to be a key component of physiopathogenesis in ACOX1 deficiency, we used the murine BV-2 microglial cell line as model: (i) to evaluate the antioxidant and anti-inflammatory properties of Opuntia ficus-indica cactus cladode extracts; (ii) to characterize a new BV-2 ACOX1 deficient cell line recently generated in our laboratory by CRISPR/Cas9 gene editing. In the first part of this work, the microglia activation, achieved by BV-2 cells exposition to four structurally and biologically well-defined lip…
Microglial and BDNF impact on the induction of the post ischemic neuroplasticity
2011
Evidences showing that under certain circumstances, inflammatory response could be neuroprotective and could also promote adult neurogenesis are growing. In this context, the objective of this work was to investigate the impact of microglial cells in the neuroplastic events. Rats were subjected to photothrombotic ischemia and microglial cells activation was blocked by the mean of poly(ADP-ribose)polymérase-1 (PARP-1) inhibition using 3- aminobenzamide (3-AB) since this protein has been shown to play a major role in this activation. Our results show that PARP-1 activity reduction was associated with a strong repression of the acute microglial activation. Beside, 3-AB treated animals exhibite…
Microglia and α-synuclein implication in Parkinson's disease
2011
Age-related neurodegenerative disorders like Parkinson’s disease take an enormous toll on individuals and on society. Despite extensive efforts, Parkinson’s disease remains incurable and only very limited treatments exist. Indeed, Parkinson’s pathogenesis is still not clear and research on its molecular mechanisms is ongoing. In this study, we focused our interest on two abnormal events occurring in Parkinson’s patients, namely α-synuclein aggregation and microglial activation. We first investigated α-synuclein and its abnormal polymerisation. For this purpose, we developed novel methods, which allowed the in vitro production of different types of α-synuclein oligomers. Using highly sensiti…
Highlighting the divertsity of cactus populations (Opuntia spp.) in Morocco and modulation of lipid metabolism by natural extracts and phytosterols f…
2015
The objective of this PhD thesis is to characterize the Moroccan germplasm of the cactus Opuntia collected from different regions in Morocco, by studying the phenologic behavior, the genetic features and physicochemical composition, along with the molecules of high therapeutic potential which may have beneficial effects on the central nervous system cells from cactus extracts (fruit, flower, seed, oil, cladodes and spine) and compared to Argan oil. This work has been conducted in fours different laboratories sharing complementary analyses skills at agronomic, biochemistry, molecular levels. Our results are presented in three parts : (i) cactus agronomic characteristics, (ii) purification an…
