Use of electron spectroscopic imaging to determine element composition of the melanin granules in the stria vascularis of the guinea pig.

Electron spectroscopic imaging (ESI) was used to analyze the element content of melanin granules in the stria vascularis seen in ultrathin sections of Spurr-embedded cochleae of the guinea pig. To determine element composition, ESI images were taken at different ionization edges, and non-specific background signals were subtracted digitally by an image processing system. The presence of calcium and nitrogen in the melanin granules could be demonstrated clearly. The calcium identified in the melanin granules was then compared with the spatial distributions of calcium binding sites after the application of an antimonate precipitation method, which was used to localize loosely bound calcium. The effect of wavelet and discrete cosine transform compression of digital radiographs on the detection of subtle proximal caries. ROC analysis.

The study compared diagnostic performances of 2 different image compression methods: JPEG (discrete cosine transform; Joint Photographic Experts Group compression standard) versus JPEG2000 (discrete wavelet transform), both at a compression ratio of 12:1, from the original uncompressed TIFF radiograph with respect to the detection of non-cavitated carious lesions. Therefore, 100 approximal surfaces of 50 tooth pairs were evaluated on the radiographs by 10 experienced observers using a 5-point confidence scale. Studies on colonic cells in the lamina propria (LP) of mice are important for understanding the cellular and immune responses in the gut, especially in inflammatory bowel diseases (such as morbus crohn and colitis ulcerosa). This protocol details a method to isolate LP cells and characterize freshly isolated cells by quality control experiments to obtain cells that can be used for further investigations. In paleontology and forensic medicine, the number of these incremental lines or annulations is used to derive the age-at-death of the individual. In the 3-d-old 2-mm root tip of Pisum sativum L. cv. Lincoln the percentage of actively proliferating cells is estimated to be 70%. The remaining cells are non-cycling and arrested with 2C and 4C DNA content in G0 and in G2Q, respectively. In this work we studied the kinetic significance of these quiescent cells, using the sorting capabilities of flow cytometry and immunofluorescence techniques to detect the proliferation marker PCNA (proliferating cell nuclear antigen) inside cells within the different cell-cycle compartments.
