Discovery of novel anti-inflammatory drug-like compounds by aligning in silico and in vivo screening: The nitroindazolinone chemotype

In this report, we propose the combination of computational methods and in vivo primary screening in zebrafish larvae and confirmatory in mice models as a novel strategy to accelerate anti-inflammatory drug discovery. Initially, a database of 1213 organic chemicals with great structural variability - 587 of them anti-inflammatory agents plus 626 compounds with other clinical uses - was divided into training and test groups. Background In marine fish species, where pelagic egg and larvae drift with ocean currents, population structure has been suggested to be maintained by larval retention due to hydrographic structuring and by homing of adult fish to natal areas. Whilst natal homing of adults has been demonstrated for anadromous and coral reef fishes, there are few documented examples of philopatric migration in temperate marine fish species. However, the organization of the complement pathway(s), as well as the functions exerted by the cloned gene products, are largely unknown. To address the issue of the presence of an inflammatory pathway in ascidians, we expressed in Escherichia coli the fragment of Ciona intestinalis C3-1 corresponding to mammalian complement C3a (rCiC3-1a) and assessed its chemotactic activity on C. intestinalis hemocytes. Abstract In this article, I provide a close reading of Season 1 of the neo-Victorian TV series Carnival Row as both an ambivalent postcolonial and neo-passing narrative. I first draw on previous criticism on postcolonial neo-Victorianism and turn-of-the-century American passing novels in order to analyze Carnival Row’s contradictory revision of imperial London through its re-imagining in a fictional city named The Burgue. I then explore the conflicting ways in which the series tackles (neo-)imperialism and colonialization, as it simultaneously criticizes and reproduces imperial ideologies and stereotypes of the racial Other.
