The endocannabinoid system controls key epileptogenic circuits in the hippocampus.

SummaryBalanced control of neuronal activity is central in maintaining function and viability of neuronal circuits. The endocannabinoid system tightly controls neuronal excitability. Here, we show that endocannabinoids directly target hippocampal glutamatergic neurons to provide protection against acute epileptiform seizures in mice. Functional CB1 cannabinoid receptors are present on glutamatergic terminals of the hippocampal formation, colocalizing with vesicular glutamate transporter 1 (VGluT1). Characterization of a Ryanodine Receptor inPeriplaneta Americana

Specific binding sites for the alkaloid ryanodine were characterized in membrane preparations from sarcoplasmatic reticulum of Periplaneta americana skeletal muscle. Binding of [3H]ryanodine was optimal at pH 8 and at CaCl2 concentration of about 300 mumol l-1. The Ca-chelating agents EGTA (100 mumol l-1) and EDTA (100 mumol l-1) abolished 95% and 90% of the [3H]ryanodine binding respectively. Preincubation with Ca2+ (100 mumol l-1) restored the ryanodine binding in presence of up to 300 mumol l-1 EGTA. Androglobin, a chimeric mammalian globin, is required for male fertility

AbstractSpermatogenesis is a highly specialised process, involving multiple dedicated pathways and regulatory check-points. Defects ultimately lead to male sub-fertility or sterility, and numerous aspects of mammalian sperm formation remain unknown. The predominant expression of the latest globin family member, androglobin (Adgb) in mammalian testis tissue prompted us to assess its physiological function in spermatogenesis. Adgb knockout mice display male infertility, reduced testis weight, impaired maturation of elongating spermatids, abnormal sperm shape and ultrastructural defects in microtubule and mitochondrial organisation. Infrequent co-existence of nitric oxide synthase and parvalbumin, calbindin and calretinin immunoreactivity in rat pontine neurons.

Neurons in the laterodorsal tegmental nucleus (LDTg), ventrolateral dorsal tegmental nucleus (LDTgV), pedunculopontine tegmental nucleus (PPTg), lateral and medial parabrachial nuclei (LPB and MPB) were immunoreactive to brain nitric oxide synthase (NOS) or isoform I. Double-labeling experiments showed that very few NOS-containing neurons in the pons were immunoreactive to any of the three calcium-binding proteins: calbindin-D 28K (CB-IR), parvalbumin (PV-IR) and calretinin (CR-IR). These findings extend our previous observation in the neocortex and suggest that a population of central NOS-containing neurons can be neurochemically characterized as CB/CR/PV deficient.

Differentiation of calcium antagonists with respect to their effects in normal and skinned taenia caeci preparations

Abstract In taenia preparations, depolarized by a K+-rich medium, Ca2+ caused contraction and cinnarizine (0.4–100 μM), trifluoperazine (2–100 μM) and verapamil (0.02–10 μM) caused concentration-dependent antagonism of Ca2+, displacing the Ca2+ log concentration-effect curve to the right and depressing the maximal response. Equieffective (IC75) antispasmogenic concentrations were selected. The antispasmogenic effects of verapamil were readily offset by removing the drug from the bathing fluid but those of the other drugs were not. The calcium antagonists (antispasmogenic IC75) were then tested for spasmolytic activity in tissues generating tension in response to the EC80 of Ca2+. Verapamil …

Forces and deformations of the abdominal wall—A mechanical and geometrical approach to the linea alba

Force-elongation responses of the human abdominal wall in the linea alba region were determined by tensile tests in which the linea alba was seen to exhibit a nonlinear elastic, anisotropic behavior as is frequently observed in soft biological tissues. In addition, the geometry of the abdominal wall was determined, based on MRI data. The geometry can be specified by principal radii of curvature in longitudinal of approximately 470 mm and in the transverse direction of about 200 mm. The determined radii agree with values found in other studies. Novel Carbon Nanotubes Rolled from 6,6,12-Graphyne: Double Dirac Points in 1D Material

Two kinds of novel carbon nanotubes, namely, (N, 0) and (0, N) 6,6,12-graphyne nanotubes (6,6,12-GNTs), are constructed by rolling up the rectangular 6,6,12-graphyne sheets along two different sides into cylinders. The mechanical and electronic properties of 6,6,12-GNTs with varied N from 3 to 20 are investigated by using density functional theory. Unlike the single-wall carbon nanotubes, the Young’s moduli of 6,6,12-GNTs do not remain constant in the case of (N, 0), but the (0, N) tubes possess almost the same one around 0.32 TPa. Molecular-dynamics study of mechanical properties of copper

Mechanical properties of copper have been studied using effective-medium theory and Molecular-Dynamics simulations. At room temperature we calculate the tensile moduli of systems that are elongated along different crystal orientations. These moduli are in very good agreement with the experimental values, the difference being less than 6%. The elastic constants obtained from simulations were also in good agreement with experiments. In addition, the point of maximum stress is found to be of the same order of magnitude as the experimental value. Blown-up toric surfaces with non-polyhedral effective cone

We construct examples of projective toric surfaces whose blow-up at a general point has a non-polyhedral pseudo-effective cone, both in characteristic $0$ and in every prime characteristic $p$. As a consequence, we prove that the pseudo-effective cone of the Grothendieck-Knudsen moduli space $\overline M_{0,n}$ of stable rational curves is not polyhedral for $n\geq 10$ in characteristic $0$ and in characteristic $p$, for all primes $p$. Many of these toric surfaces are related to a very interesting class of arithmetic threefolds that we call arithmetic elliptic pairs of infinite order.
