In vitro analysis of the phenotypical and functional properties of the 4F7+ cutaneous accessory dendritic cell

The monoclonal antibody 4F7 detects a molecule on dermal and epidermal Ia+ dendritic cells (DCs), and some of these cells are Birbeck granule-containing cells. Here we report on the phenotypical and functional characteristics of these cells which were highly enriched by 4F7-labelled immunomagnetic beads. The ultrastructural, immunocytochemical and cytochemical analyses of these preparations showed cells with the typical characteristics of DCs. The cells were found to express the DC marker NLDC145, but not 33D1. The C3bi receptor and marker F4/80 were only expressed by epidermal 4F7+ cells. In the present study, the pattern of some extracellular matrix glycoproteins was determined in traumatic anterior cruciate ligament ruptures (n = 17) and in normal ligaments (n = 11). 6 microns cryo-sections were incubated with monoclonal antibodies recognizing tenascin, laminin, and fibronectin, and FITC-labeled secondary antibodies. Immunohistochemistry revealed that tenascin, fibronectin and laminin are sparsely distributed in normal anterior cruciate ligaments, but strongly expressed in ruptured ligaments with a time-restricted pattern. Immunoreactivity of monoclonal anti-melanoma antibodies in relation to the amount of radioactive iodine substituted to the antibody molecule.

The damage to monoclonal anti-melanoma antibodies caused by iodination was investigated by comparing the results obtained using the chloramine-T method and the 1,3,4,6-tetrachloro-3 alpha, 6 alpha-diphenyl-glycoluril (IODOGEN) method at different levels of iodine substitution to the molecule. The level of substitution at which losses in immunoreactivity occurred was evaluated in each monoclonal antibody (MAb) studied. This phenomenon was not dependent on the method of substitution, provided that mild conditions of reaction were used.
