Oligonucleotides-decorated-poly(N-vinyl pyrrolidone) nanogels for gene delivery

Pulsed electron-beam irradiation of a semi-dilute poly(N-vinyl pyrrolidone) (PVP) aqueous solution in the presence of acrylic acid has led to a carboxyl functionalized nanogel system. Nanoparticles hydrodynamic size and surface charge density, in water and as a function of pH, were investigated by dynamic light scattering and laser doppler velocimetry, respectively. Nanogels (NGs) were proved not to be cytotoxic at the cellular level. A growing body of evidence shows that Insulin, Insulin Receptor (IR) and IR signaling are involved in brain cognitive functions and their dysfunction is implicated in Alzheimer's disease (AD) neurodegeneration. Thus, administration of insulin could be a strategy for AD treatment. En esta tesis doctoral se muestra la conversión directa de fibroblastos embrionarios de ratón a células con morfología y fenotipo similares al hepatocito mediante la expresión retroviral de seis factores de transcripción hepáticos, Hhex, Gata4, Foxa2, HNF4α HNF1α y HNF6. Los fibroblastos infectados mantenidos durante 3-4 semanas en cultivo adquieren una morfología epitelial y expresan genes típicamente hepáticos como Albumina, Cyp7a1, Tat o Slc10a1 a niveles próximos al hepatocito.
