Functional stability of the nitrate-reducing community in grassland soils towards high nitrate supply

Abstract To study the effects of short-term fluctuation of nitrate concentrations on the nitrate-reducing community, repacked soil cores were amended with 0, 100 and 300 μ g NO 3 - ‐ N g - 1 soil and incubated for 3, 7 and 14 days, respectively. The nitrate reductase activity was determined in a laboratory-based enzyme assay. In parallel, the community structure of nitrate-reducing microorganisms was characterised by RFLP-PCR using the functional gene narG , which encodes the catalytic site of the membrane-bound nitrate reductase. We determined the concentration of nitric oxide metabolites (NO2-+NO3-), expressed as NOx, in several clinical conditions. A kinetic study of the basic hydrolysis of 2-phenylethyl nitrite in the presence of borate buffer and β-cyclodextrin

The kinetics of the hydrolysis of 2-phenylethyl nitrite have been studied in basic aqueous solutions (borate buffer) in the absence and presence of β-cyclodextrin. The observed kinetic trends show that the hydrolysis reaction consists of at least three subsequent processes. The first is a fast acid-base reaction between the boron atom of boric acid and oxygen atom of the alkyl nitrite. The second one should be the nucleophilic attack of hydroxyl ion on the alkyl nitrite. The last process could be referred to as a combination of transfer of hydrogen acid to a water molecule followed by ruptures of nitrogen-oxygen and boron-oxygen bonds. Some observations on the spectrophotometric determination of nitrite using ergonovine-p-dimethylaminobenzaldehyde

Abstract The system ergonovine maleate-p-diethylaminobenzaldehyde in sulfuric acid media is applied to the kinetic-spectrophotometric determination of nitrite ions with competitive precision. The reaction rate is strongly enhanced by the presence of nitrite or ferric ions, being the nitrite in the “promoting” role of the reaction. Linear calibration plots are obtained for nitrite concentration ranges 0.3–2.4 ppm or 0.66–6 ppb. The influence of foreign compounds is also established.

Janus-faced role of endothelial NO synthase in vascular disease: uncoupling of oxygen reduction from NO synthesis and its pharmacological reversal

Endothelial NO synthase (eNOS) is the predominant enzyme responsible for vascular NO synthesis. A functional eNOS transfers electrons from NADPH to its heme center, where L-arginine is oxidized to L-citrulline and NO. Common conditions predisposing to atherosclerosis, such as hypertension, hypercholesterolemia, diabetes mellitus and smoking, are associated with enhanced production of reactive oxygen species (ROS) and reduced amounts of bioactive NO in the vessel wall.
