Mutations in the NKX2.5 Gene and the PAX8 Promoter in a Girl with Thyroid Dysgenesis

Screening of the known candidate genes involved in thyroid organogenesis has revealed mutations in a small subset of patients with congenital hypothyroidism due to thyroid dysgenesis (TD).We studied a girl with TD who had mutations in two transcription factors involved in thyroid development.Sequencing analysis of candidate genes involved in thyroid gland development revealed a new paternally inherited heterozygous mutation in the NKX2.5 gene (S265R) and a new maternally inherited heterozygous mutation in the PAX8 promoter region (-456CT). Publisher Summary This chapter discusses the temporal development of early and late contingent negative variation (CNV) in a simple discrimination paradigm. The CNV typically arises when a warning stimulus is paired with an imperative stimulus which, in turn, is followed by a motor reaction. If the interstimulus interval is less than approximately 2 sec the CNV begins immediately after the positive deflection (P300) following the warning stimulus. It may appear as a steadily rising negative potential shift that reaches its peak with the presentation of the imperative stimulus, or as sustained negativity throughout the rest of the foreperiod. The notion that some individuals are more prone to emotion transmission than others has prompted the need for a person-oriented approach to emotion transmission in parent-child dyads. The present study applied a person-oriented analysis to examine the patterns of emotion transmission that can be identified in the diary data of father-child dyads, and the extent to which children with high levels of temperamental negative emotionality are particularly susceptible to emotion transmission within the family.
